AB - In the context of the macroscopic quantum phenomena of the second kind, we hereby seek for a solution-in-principle of the long standing problem of the polymer folding, which was considered by Levinthal as (semi)classically intractable. To illuminate it, we applied quantum-chemical and quantum decoherence approaches to conformational transitions. Our analyses imply the existence of novel macroscopic quantum biomolecular phenomena, with biomolecular chain folding in an open environment considered as a subtle interplay between energy and conformation eigenstates of this biomolecule, governed by quantum-chemical and quantum decoherence laws. On the other hand, within an open biological cell, a system of all identical (noninteracting and dynamically noncoupled) biomolecular proteins might be considered as corresponding spatial quantum ensemble of these identical biomolecular processors, providing spatially distributed quantum solution to a single corresponding biomolecular chain folding, whose density of conformational states might be represented as Hopfield-like quantum-holographic associative neural network too (providing an equivalent global quantum-informational alternative to standard molecular-biology local biochemical approach in biomolecules and cells and higher hierarchical levels of organism, as well).

AB - The complex phenomenon of rearrangement of band 3 molecules after erythrocyte swelling under hypotonic condition is considered. The rearrangement includes the increase of the mobile fraction and clustering of band 3. The self-associative tendency and the action of the elastic field generated within the lipid membrane after erythrocyte swelling result in equilibration of the number of molecules per cluster and the number of clusters. The local perturbation of the elastic field induces excitation of the cluster in the nearest neighbor and changes its packing state generating changes in the free volume within the cluster. The local perturbation could result in the reversible formation of osmotic hole. We formulated a model to predict changes of the cluster packing states generated by rearrangement of band 3 molecules on two time-scales. The phenomenon is examined on the basis of two experimental sets, i.e. low (5.2 mM Na3PO4 solution) and high (46.0 mM Na3PO4 solution) hypotonicities at 21A degrees C, from Golan and Veatch (Proc Natl Acad Sci 77(5):2537-2541, 1980). Modeling considerations suggested that lower hypotonic conditions resulted in higher values of: the driving force of agglomeration of band 3 as a measure of self-associative tendency, the specific rate of cluster breaking, the specific rate of increase of the mobile fraction of band 3, and the dispersion of cluster sizes. Lower hypotonic conditions ensure the generation of a higher average value of the free energy within the membrane after erythrocyte swelling, which enables more intensive rearrangement of band 3 molecules.

abstract = "The complex phenomenon of rearrangement of band 3 molecules after erythrocyte swelling under hypotonic condition is considered. Lower hypotonic conditions ensure the generation of a higher average value of the free energy within the membrane after erythrocyte swelling, which enables more intensive rearrangement of band 3 molecules.",

AB - Interaction of cells with polymer networks in nanostructured devices as alginate beads for cell immobilization are considered. Influence of process parameters as oxygen concentration inside a bead on changes of alginate material structure is experimentally investigated. Significant swelling of alginate beads during process is observed and ascribed to combined action of chemical components in the reactor and of growing cells, causing expansion of beads and degradation of their networks. The culture of mouse/mouse hybridoma cells secreting a monoclonal antibody (IgG) specific to human chorionic gonadotropin (beta-hCG) was used. A kinetic model of catalytic alginate degradation is developed for controlling the process. The model provides possibilities to analyze conditions for suppressing large swelling and influence of oxidative agents. Moreover, it provides some deeper insights into mechanism of polymer network-cell interactions and dynamics during the process. It is based on separation of contributions of reactants in the system from contributions of cells to degradation of the network and in that way to effective swelling of the bead.

AB - A new approach to the two-level design and self-assembling of MexSiOy (Me = Gd, Y and Ca or their mixtures) nano-powders from SiO2 sols is described. The nanostructure design of various optical active materials based on different compounds of Me(x)Sio(y) is discussed. These particles were formed by the combination of hydrothermal synthesis and ultrasound spray pyrolysis. The morphology and structural design of the particles and sub-particles are defined by their synthesis procedure and processing parameters. The mean particle size and size distribution of all the MexSiOy based compounds were determined by SEM analysis. These results were compared with the particle diameter evaluated from the theoretical model of the breaking-up of capillary waves. For all the synthesized powders, the obtained results indicate the possibility of a rigorous design of the structure of the particles throughout their entire hierarchy.

AB - Aim. A phase-field mathematical model was formulated to describe yeast cell growth within the Ca-alginate microbead during air-lift bioreactor cultivation. Model development was based on experimentally obtained data for the intra-bead yeast cell volume fraction profile within the microbead after reaching the equilibrium state for cells (150 h), as well as, total yeast cell volume fraction per microbead and microbead volume as functions time. Microbead with growing yeast cells is treated as a two-phase system. one phase represents the cell agglomerates, while the other is the alginate hydrogel matrices. The interactions between phases are simulated using the Langevin class, non-conservative phasefield model based on the reduction of the modeling resolution. The model considered the growth of small domains of one phase (cell agglomerates) as nucleation. Methods. Total yeast cell volume fraction in the beads was estimated by using Thoma counting chamber after dissolution of beads. Local cell volume fraction per microbead layers is calculated from experimentally determined surface fraction of cells for various microbead cross sections by image analysis. Microbead volume is estimated by measuring the microbead diameter. Diameters of microbeads were measured using the optical microscope equipped with a micrometric device. Results. The proposed model offered the only one model parameter, which represents the specific measure of microenvironmental restrictive action to the cell growth dynamics. The optimal value of this model parameter is obtained by comparison analysis between experimental data and model predictions. Conclusion. Besides giving useful insights into the dynamics of restrictive cell growth within the Ca-alginate microbead, the model can be used as a tool to design/optimize the performance of microbead and studying the microenvironmental restrictive mechanism action of the cell growth.

AB - A mathematical model was formulated to describe hybridoma cell growth within the alginate-poly-L-lysine (alginate-PLL) microcapsules during air-lift bioreactor cultivation. Model development was based on experimentally obtained data concerning the hybridoma cell counts, monoclonal antibody (mAb) production and the distribution of hybridoma cell growth within the microcapsules. The cell growth was modeled using a mean field approach expressed as Langevin class of equations for two different regions of alginate-PLL microcapsules, the alginate microcapsule core and the annular region between microcapsule core and membrane. In this paper we propose an influence of microenvironmental conditions on cell growth. The osmotic pressure changes in the Na-alginate liquefied annular region, as well as, the resistance effects of Ca-alginate hydrogel in the core region during the cell growth were incorporated into the model. Good agreement between the experimental data and model prediction values was obtained. The proposed model successfully predicted the impact of various microenvironmental restriction effects on the dynamics of cell growth and appears useful for further optimization of microcapsule design in order to achieve higher intra-capsule cell concentrations resulting in higher amounts of mAb produced.

Model development was based on experimentally obtained data for the intra-bead cell concentration profile, after reached the equilibrium state, as well as, total yeast cell concentration per microbed and microbead volume as function of time. Relatively uniform cell concentration in the carrier matrix indicated that no internal nutrient diffusion limitations, but microenvironmental restriction, affected dominantly the dynamics of cell growth. Also interesting phenomenon of very different rates of cell number growth during cultivation is observed. After some critical time, the growth rate of cell colonies decreased drastically, but than suddenly increased again under all other experimental condition been the same. It is interpreted as disintegration of gel network and opening new free space for growth of cell clusters. These complex phenomena are modeled using the thermodynamical, free energy formalism. The particular form of free energy functional is proposed to describe various kinds of interactions, which affected the dynamics of cell growth and cause pseudo-phase transition of hydrogel. The good agreement of experimentally obtained data and model predictions are obtained. In that way the model provides both, the quantitative tools for further technological optimization of the process and deeper insight into dynamics of cell growth mechanism.

AB - The rheological model is developed to elucidate the mechanism of Ca-alginate microbead deformation in the course of cell growth within. It is a complex process influenced by relaxation of the expanded polymer network inside a bead, and forces generated by cell growth inside the bead and interactions between solvent, network parts and cells as well. The resulting effects are measured experimentally by estimating isotropic volumetric deformations of beads with yeast cells as function of time and cell concentration per bead. The mathematical model of the process is developed based on a modified general Zener model with fractional derivatives. It is particularly suitable for incorporating effects of different nature also during different stages of such complex process development. The results of theoretical analyses using the model developed and comparison with experimental values obtained, indicate a high impact of partial decomposition, i.e. plastic response of polymer network inside a bead due to cell growth, on bead deformation. For comparison, corresponding deformation measurements and modelling were performed on the same network system exposed to swelling in the solvent, but without the cells. In this case elastic forces are dominant, indicating different mechanism of relaxation without the influence of cells, in agreement with previous conclusions.

AB - A phase-field model was formulated to describe yeast cell growth within the Ca-alginate microbead during air-lift bioreactor cultivation. Model development was based on experimentally obtained data for intra-bead yeast cell volume fraction profile after reaching the equilibrium state, as well as, total yeast cell volume fraction per microbead and microbead volume as functions of time. Relatively uniform cell concentration in the microbead, experimentally obtained, indicated microenvironmental restriction effects due to deformation and disintegration of previously swollen alginate hydrogel, while internal nutrient diffusion limitation could be neglected in the case of alginate microbeads used in this study. Microbead with growing yeast cells is treated as two phase system. One phase represents the cell agglomerates, while the other is the alginate hydrogel matrices. The interactions between phases are modeled using the Langevin class, non-conservative phase-field model. Such class of models is suitable for considering the growth of small domains of one phase as nucleation. Besides giving useful insights into the dynamics of restrictive cell growth within the Ca-alginate microbead, the model can be used as a tool to design/optimize the performance of microbead and studying the microenvironmental restrictive mechanism action on the cell growth. .

AB - The development of lipid-based fine particles as potential drug carriers requires a detailed investigation of the possible effects of these carriers on the rheological properties of blood. In this study, we investigated the influence of dynamic conditions on aggregate formation and stability in dispersions of lipid-based fine particles in whole blood under in vitro conditions. The rheological parameters of two concentrations of liposome dispersion and two concentrations of lipid emulsion in blood were examined. A micro-rheological model of aggregating dispersions is proposed in which the apparent viscosity is estimated as the sum of the hydrodynamic and structural parts which are correlated with system structural ordering in the flow. The dynamics of structural ordering of the aggregating system an considered by examining the evolution of the state of the system in phase space depending on the shear rate. The addition of lipid-based particles induced aggregate formation in the blood, which was more pronounced at higher concentrations of lipid-based fine particles. Furthermore, larger and more stable aggregates are formed in liposome dispersions as compared to lipid emulsions in blood.

The aggregation of phospholipid vesicles (liposomes) in water is analyzed experimentally and theoretically and compared with the well-examined alumina dispersions. The steady-state aggregation followed rheologically for various volume fractions of particles and shear rates. Methods. A micro-rheological model of aggregating dispersions is proposed in which the apparent viscosity is estimated as the sum of hydrodynamic and structural parts, which are correlated with system structural ordering. Langevin equation is used to describe the dynamics of evolution of steady-states of liposome systems in phase space, for various shear rates. Results. These results established a link between the local dynamics of liposome dispersions and their flow behavior. Conclusion. Aggregates become more compact and embedded higher volume of fluid when shear rate increases. Hydrodynamic part of viscosity increases, while structural part of viscosity decreases with shear rate. Calculated value of bond energy between liposomes corresponds to hydrogen bond energy.

AB - Design of mechanical properties, especially the relation between tensile and dynamic moduli in terms of chain conformational statistics is considered. Data on new elastomer blends with gradient properties, able for mixing on molecular level and partial networking with separation of phases, are analyzed in parallel to classical commodity materials. Especial attention is paid to synergetic effects of the increase of blend modulus, relative to moduli obtained by linear "rule of mixtures" for component polymer moduli, described by Kleiner-Karasz-MacKnight equation, and new models based on self-similar scaling of elastomer network dynamics. New data on model blends exposing synergetic effects with the change of conformational statistics are presented as well.

abstract = "Design of mechanical properties, especially the relation between tensile and dynamic moduli in terms of chain conformational statistics is considered. New data on model blends exposing synergetic effects with the change of conformational statistics are presented as well.",

AB - The change in elastomer tensile moduli, as formulated in the Gaussian statistical theory of rubber elasticity, with deformation, is considered both experimentally and theoretically. Gum elastomers of different structures and corresponding materials filled with carbon black, as reinforcing filler, are investigated experimentally. For all materials considered, the same scaling pattern with negative and low slope for small deformations, and positive and higher slope for large deformations is obtained, indicating two distinct mechanisms of elastic response. Most pronounced is the similarity of small deformation responses for filled materials. Considering the modulus as an elastic energy density gradient dependent on structure changes with deformation, and interpreting the changes for small deformations in terms of conformational energy change, the fractal dimension of a new type is formulated. It describes the decrease in elastomer network connectivity with deformations, which is discussed in terms of conformon dynamics. Possibilities of application of Faynman's path integral method and statistical method of random walk to the lattice are considered for the conformon, as well.

The specific rate of elastic modulus increase, as well as the specific rate of loss modulus increase were used as a measure of chains flexibility in the flow field. The results were compared with the theory of Rouse proposed for flexible chains. All values of the specific rate of elastic modulus increase and the specific rate of loss modulus increase were higher then value from Rouse theory, indicating specific scaling and high rigidity of Na-alginate chains.

AB - The relations between the structure and mechanical properties of a polymer blend of natural (NR) and polybutadiene (BR) rubber (i.e. a NR/BR blend with the weight ratio of the components 70/30) filled with active carbon black were analysed. The properties of the individual phases in the blend were resolved by modeling the stress-strain relationship according to the Bauer procedure for high extensions. The obtained results indicated that BR is the dispersed phase, having a higher modulus, which was also confirmed by the much better fit of the experimental data to the series type of phase coupling according to the Takanayagy theory.

Svojstva individualnih faza u 70/30 NR/BR blendi mogu se na zadovoljavajući način definisati modelom koji uzima u obzir zavisnost napon-deformacija pri velikim istezanjima. AB - Development of lipid-based fine particles as potential drug carriers requires detailed investigation of possible effects of these carriers on rheological properties of blood. In this study, we have investigated the influence of dynamic conditions on aggregate formation and stability in dispersions of lipid-based fine particles in whole blood under in vitro conditions. Rheological parameters of two concentrations of liposome dispersion and two concentrations of lipid emulsion in blood were studied by assessing shear stress/shear rate relationships. The magnitude of attractive interactions between aggregates and/or particles, A, and the effective-to-real volume fraction of particles, phi(f)/phi(p) , were estimated for rheological quantification of lipid-based fine particles-blood interactions and aggregate stability. Addition of lipid-based particles induced aggregate formation in blood, which was more pronounced at higher concentrations of lipid-based fine particles. Furthermore, larger and more stable aggregates were formed in liposome dispersions as compared to lipid emulsions in blood.

AB - A new procedure for introduction of aromatic diamines into polymer, in particular into poly(vinyl chloride) (PVC) molecule, was developed. Produced materials have attractive properties such as combination of good film-forming, coating and adhesive features (especially adhesion to hard-to-adhere substrates, as glass) and functionalization possibilities. Free pendent amine groups enable nucleophilic additions with fullerene molecule C-60, for creating especial properties of such a film.

AB - Chemically synthesized hydroxyapatite/poly-L-lactide (HAp/PLLA) composite biomaterial was studied in vivo. The biocomposite was implanted into Balb/c Singen mice and after 1 and 3 weeks removed from their organisms and analyzed by the FT-IR spectroscopy. After 1 week of testing in vivo the implanted sample gave a spectrum in which absorption bands arising from newly formed functional groups of amine and peptide can be seen. After 3 weeks, a spectrum with pronounced absorption bands at 3420 and 1650cm-1 assigned to newly generated collagen, a component of the extracellular connective-tissue matrix, was registered. Also, decrease of the intensity absorption band at 1760cm-1 originating from the C=O group of PLLA indicates bioresorption of the PLLA used. Analysis of the microstructure of the sample surface by scanning electron microscopy before and after implantation revealed bioresorption of the PLLA polymer phase and generation of collagen fibers at the sites of implanted bioresorptive PLLA. A mixture of autologous bone powder and HAp/PLLA biocomposite was also examined. After implantation, the same final products as in the case of HAp/PLLA composite biomaterial used alone were found. abstract = "Chemically synthesized hydroxyapatite/poly-L-lactide (HAp/PLLA) composite biomaterial was studied in vivo.
