AB  - A priori estimation of analyte response is crucial for the efficient development of liquid chromatography–electrospray ionization/mass spectrometry (LC–ESI/MS) methods, but remains a demanding task given the lack of knowledge about the factors affecting the experimental outcome. In this research, we address the challenge of discovering the interactive relationship between signal response and structural properties, method parameters and solvent-related descriptors throughout an approach featuring quantitative structure–property relationship (QSPR) and design of experiments (DoE). To systematically investigate the experimental domain within which QSPR prediction should be undertaken, we varied LC and instrumental factors according to the Box-Behnken DoE scheme. Seven compounds, including aripiprazole and its impurities, were subjected to 57 different experimental conditions, resulting in 399 LC–ESI/MS data endpoints. To obtain a more standard distribution of the measured response, the peak areas were log-transformed before modeling. QSPR predictions were made using features selected by Genetic Algorithm (GA) and providing Gradient Boosted Trees (GBT) with training data. Proposed model showed satisfactory performance on test data with a RMSEP of 1.57 % and a of 96.48 %. This is the first QSPR study in LC–ESI/MS that provided a holistic overview of the analyte's response behavior across the experimental and chemical space. Since intramolecular electronic effects and molecular size were given great importance, the GA–GBT model improved the understanding of signal response generation of model compounds. It also highlighted the need to fine-tune the parameters affecting desolvation and droplet charging efficiency.

Impact of wastewater antibiotics on river water quality in Belgrade area

AB  - Municipal wastewater in the Belgrade area is discharged into the Danube and the Sava
Rivers without any treatment. This affects the quality of river water since wastewater is one of the
most important sources of environmental pollutants, including pharmaceuticals. To assess the impact
of this effect, the presence of three commonly used antibiotics in wastewater and receiving river water
was determined. A previously developed liquid chromatography-tandem mass spectrometry method
for the analysis of antibiotics in natural water matrices was validated for wastewater and applied in
analysis of samples from the Belgrade area. All investigated antibiotics were detected in both
wastewater and river water samples. The highest concentration of antibiotics in wastewater was
found for sulfamethoxazole, while trimethoprim was detected at the highest level in surface water.
The most frequently detected antibiotic in surface water was azithromycin. The environmental risk
assessment was performed for surface water biota. Azithromycin was found to pose a high risk to
aquatic organisms, while the risk level of sulfamethoxazole was medium or low. In conclusion, the
obtained results show the detrimental impact of untreated wastewater discharge on the river water
quality in the Belgrade area, emphasizing the need for urban wastewater treatment.

AB  - Glavni put dospevanja lekova u životnu sredinu su komunalne otpadne vode. U regionu
Beograda, otpadne vode se direktno ispuštaju u reke Savu i Dunav, što negativno utiče na kvalitet
rečne vode. Zbog svoje postojanosti i biološke aktivnosti, lekovi mogu imati veoma štetan uticaj na
živi svet u vodenoj sredini. U ovoj studiji je ispitivano prisustvo pet najčešće korišćenih
kardiovaskularnih lekova u rečnoj vodi Beograda, kao i u odgovarajućim otpadnim vodama. Za
ekstrakciju uzoraka vode korišćena je prethodno razvijena metoda za određivanje lekova u
različitim vodenim matricama. Dobijeni ekstrakti su analizirani metodom tečne hromatografije u
sprezi sa tandem masenom spektrometrijom, uz korišćenje elektrosprej jonizacione tehnike.
Rezultati su pokazali da su najčešće detektovani kardiovaskularni lekovi i u površinskim i u
otpadnim vodama bili metoprolol i bisoprolol, koji se na osnovu dosadašnjih ekotoksikoloških
istraživanja smatraju štetnim za vodene organizme.

AB  - Veštački zaslađivači su sintetičke supstance koje se koriste za postizanje slatkog ukusa prehrambenih proizvoda, a čija je upotreba u Republici Srbiji regulisana Pravilnikom o prehrambenim aditivima. Nakon ingestije, najveći procenat ovih jedinjenja prolazi u neizmenjenoj formi kroz ljudsko telo, i kao takva dospevaju u životnu sredinu putem komunalnih otpadnih voda. Naučna istraživanja u poslednjoj deceniji su pokazala široku rasprostranjenost veštačkih zaslađivača u površinskim i podzemnim vodama. Zbog ograničenih podataka o njihovoj toksičnosti po živi svet u vodi, kao i o ponašanju u akvatičnim ekosistemima, svrstani su u klasu emergentnih zagađujućih materija.
Ovaj rad opisuje analizu široko korišćenog zaslađivača ciklamata metodom tečne hromatografije u sprezi sa tandem masenom spektrometrijom u netretiranim otpadnim vodama Beograda radi procene prisustva i unosa ovog jedinjenja u životnu sredinu. Rezultati dobijeni analizom četiri uzorka otpadnih voda iz različitih kanalizacionih ispusta u Beogradu pokazuju prisustvo ciklamata u opsegu koncentracija od 4,3 µg L–1 do 23,2 µg L–1. S obzirom na to da Beograd ne poseduje postrojenje za preradu otpadnih voda, visoki nivoi detektovanog ciklamata ukazuju na kontinualnu kontaminaciju vodenih ekosistema u Republici Srbiji i potencijalnu opasnost po živi svet u vodi.
AB  - Artificial sweeteners are synthetic substances that are used to achieve the sweet taste of
food products, and their application in the Republic of Serbia is regulated by the national
guidelines on food additives. After ingestion, the largest percentage of these compounds passes
unchanged through the human body and reaches the environment through municipal wastewater. In
the last decade, studies have shown the widespread use of artificial sweeteners in surface and
groundwater. Due to limited data on their toxicity to wildlife in the water, as well as on their
behaviour in aquatic ecosystems, they are classified as emerging contaminants.
This paper describes the analysis of widely used sweetener cyclamate by liquid chromatography
with tandem mass spectrometry in untreated urban wastewater of Belgrade to assess the presence
and input of this compound into the environment. The results obtained by analysing four
wastewater samples from different sewage channels in Belgrade show the presence of cyclamate in
the concentration range of 4.3 µg L
–1
–23.2 µg L–1
. Since Belgrade does not have a wastewater
treatment plant, high levels of cyclamate indicate the continuous contamination of aquatic
ecosystems in Serbia and the potential danger to wildlife in the water.

AB  - In the city of Belgrade, raw sewage is discharged directly into the Sava and the Danube rivers affecting the quality of river water. To determine the level of this impact, eight frequently used and detected pharmaceuticals were selected for the study. A previously developed method for analysis of pharmaceuticals in different water matrices was used. Sample extracts were analyzed by liquid chromatography-tandem mass spectrometry, using the electrospray ionization technique. The most commonly detected analytes in both municipal wastewater and surface water were carbamazepine, known for its high environmental stability, and diclofenac, known as having high acute ecotoxicity. The dilution effect, noted for most of the detected drugs, was not as pronounced with diclofenac, indicating a higher risk for aquatic organisms.

AB  - Veštački zaslađivači se decenijama koriste kao aditivi u hrani, piću i farmaceutskim proizvodima. Savremena istraživanja su pokazala široku rasprostranjenost ovih supstanci u prirodnim i otpadnim vodama, zbog čega su svrstani u klasu emergentnih zagađujućih materija. U ovom radu je prikazana optimizacija metode pripreme uzoraka vode ekstrakcijom na čvrstoj fazi za određivanje tragova odabranih veštačkih zaslađivača. Ova metoda omogućava istovremeno izolovanje i koncentrovanje analita, ali i prečišćavanje ekstrakta uzorka. Prilikom razvoja metode izvršen je izbor optimalnog adsorbenta, zapremine i pH vrednosti uzorka vode, kao i rastvarača za eluiranje analita sa pakovanja kolone. Dobijeni ekstrakti su analizirani metodom tečne hromatografije–masene spektrometrije.

AB  - The importance of studying electrospray ionization (ESI) technique stems from its widespread liquid
chromatography – mass spectrometry (LC-MS) character, particularly emphasized in scientific areas with most
of non-volatile and highly- or semi- polar analytes. Although it is known that ESI operating gas-phase ions are
formed under the influence of electrical – thermal – pneumatic energy, applied to analyte-containing liquid after
its removal from capillary tube, insufficiently explained complexity of process decelerates finding optimal
instrumental settings and, hence, providing high ionization efficiency. Special consequence represents obtaining
varying responsiveness among group of analytes of identical concentration levels, measured under the same
experimental conditions. In this regard, developing quantitative analytical methods could be compromised.
Taking into account previously associated findings, the purpose of this study was to examine LC-MS response
behavior of test substances - atypical antipsychotic aripiprazole and its impurities in dependence with
instrumental, solvent and analyte-related properties by employing Quantitative Structure – Property Relationship
(QSPR) approach. QSPR methodology primarily quantifies correlation between property of interest, determined
for a series of analytes in a chosen analytical system, and molecular descriptors - in silico calculated or
experimentally obtained numerical values attributed to certain chemical information. Assuming nonlinear
relationship between observed variables, artificial neural network (ANN) was used for establishing a
comprehensive QSPR model with good response prediction ability. Data table for model building was composed
of molecular descriptors, as well as, LC and ESI source parameters varied according to Box-Behnken design of
experiments. Set of molecular descriptors included polar surface area, pKa, logP, logD, molecular volume,
surface tension, vapor pressure, number of proton acceptors of analytes, as well as, viscosity, conductivity and
surface tension of utilized mobile phases, due to encoding ESI-relevant chemical information. Additionally, the
amount of methanol in mobile phase, pH of aqueous portion, flow rate of mobile phase, sheath gas flow,
auxiliary gas flow, spray voltage and capillary temperature were considered as model’s input variables, based on
significant impact on peak areas, shown within previously performed Placket-Burman design. Within it,
unexpectedly occurred confounding patterns, guided by the alias matrix methodology, directed settings of
involved, but statistically insignificant screened factors.
Predictive power of finally obtained model was estimated using internal, 10-fold cross-validation procedure.
Developed QSPR model showed satisfactory performance in terms of low root mean square errors (RMSE) and
relatively high values of cross-validated coefficient of determination (Q2). In particular, study has demonstrated
suitability of utilized modeling approach in finding the optimal experimental parameters and, hence, usefulness
for efficient LC-MS method development. However, generalization of manifested underlying physicochemical
mechanisms must be precluded and limited only to the examined system, regarding relatively small number of
available structures.

AB  - Elektrosprej	jonizacija	 (ESI)	predstavlja	najčešće	korišćenu	 tehniku	jonizacije	u	
LC/MS	analizi	polarnih	i	umereno	polarnih	analita.	Nedovoljno	 rasvetljeni	mehanizmi	
generisanja	ESI	jona	uslovljavaju	dugotrajnu	optimizaciju	odgovora	sistema,	zasnovanu	
na	 primeni	 pristupa	 pokušaja‐i‐greške.	 Upotreba	 metodologije	 kvantifikovanja	 veze	
strukture	analita	sa	osobinom	od	interesa	 (QSPR),	odnosno,	ESI	signalom	može	da	dâ

AB  - After decades of using artificial sweeteners in the food and pharmaceutical industry, their ubiquitous presence in the environment has been determined 1,2. These substances are recognized as emerging pollutants due to limited data on their occurrence, environmental fate and ecotoxicity. Prior to analysis of artificial sweeteners at trace levels in water samples, it is necessary to isolate and concentrate analytes and clean-up the obtained extracts. In this paper, the optimization of the preparation method for the determination of saccharin, sucralose and neotame in water by solid-phase extraction (SPE) was performed. The extracts were analyzed by liquid chromatography–tandem mass spectrometry (LC–MS/MS). In the SPE optimization, the following parameters were tested: the sorbent type, the pH value and the volume of water sample and the elution solvent. The suitability of anion-exchange, polymeric, C18 and carbon sorbents for the extraction of selected sweeteners was assessed. The results showed that the highest recoveries were obtained using polymeric Oasis HLB cartridges, with copolymer poly(divinylbenzene-co-N-vinylpyrrolidone). Furthermore, it was determined that the pH value of water sample was crucial for extraction efficiency. For all three artificial sweeteners, the optimal pH value of the sample was 3. Finally, the sample volume of 100 ml ensured a high preconcentration factor without the sorbent breakthrough, and methanol was selected as the elution solvent.

AB  - Multifunctional drug anisomycin was subjected to forced degradation in accordance with International Conference on Harmonisation (ICH) guidelines for the first time. The drug was exposed to the recommended stress conditions of hydrolysis (acidic, alkaline and neutral), oxidation, thennal stress and photolysis, in order to investigate its stability. Optimized LC-MS/MS method was validated as recommended by ICH Q2(R1) guideline with respect to the specificity, accuracy, precision, limits of detection and quantitation, linearity and robustness. Anisomycin exhibited high instability under alkaline and thermal (neutral hydrolysis) conditions. It showed moderate stability under acidic, neutral, oxidative, thermal (acidic hydrolysis) and photolytic conditions, with the lowest degradation level observed in the case of light and oxidation stress. Formation of the same degradation product, identified as deacetylanisomycin, was observed under all applied stress conditions.

Pregled regulativa usmerenih na emergentne zagađujuće materije u vodenoj sredini

AB  - Emergentne zagađujuće materije obuhvataju širok spektar supstanci koje mogu imati
potencijalno štetno dejstvo na zdravlje ljudi. Takođe, usled velike rasprostranjenosti i postojanosti
u životnoj sredini, pokazano je da ove supstance, čak i pri niskim koncentracijama, dovode do niza
neželjenih uticaja na živi svet, u najvećoj meri kod vodenih organizama. U životnu sredinu
dospevaju iz raznih antropogenih izvora zagađenja. Emergentne zagađujuće materije nisu
uključene u redovne programe monitoringa zagađujućih materija i njihovo ponašanje i sudbina u
životnoj sredini, kao i ekotoksikološki efekti nisu u potpunosti poznati. Iz tog razloga, neophodno ih
je uvrstiti u postojeće regulative usmerene na kvalitet životne sredine.
U ovom radu urađena je detaljna analiza propisa vezanih za zagađenje vodene sredine, a koji
predstavljaju osnovu za održivo upravljanje vodnim resursima. Obrađena je zakonska regulativa
EU koja obuhvata Okvirnu direktivu o vodama, „watch listu” zagađujućih materija i Direktivu o
registraciji, evaluaciji, autorizaciji i restrikcijama hemikalija. Takođe su analizirana uputstva
Agencije za zaštitu životne sredine SAD-a za praćenje kvaliteta vode i zaštitu vodenih organizama,
kao i standardi serije ISO 14000 za menadžment životnom sredinom i standardi ISO 24510, 24511 i 24512 za usluge vodosnabdevanja i tretmana otpadnih voda. Prikazan je i koncept menadžmenta
emergentnim materijama u akvatičnoj sredini.
AB  - Emerging contaminants cover a wide range of substances which can be potentially harmful to human health. Also, it was determined that these substances due to widespread occurrence and persistence in the environment cause a series of adverse environmental effects, mostly to aquatic organisms, even at low concentrations. These contaminants reach the environment from various anthropogenic pollution sources. Emerging contaminants are not included in routine monitoring pollutant programs and their behavior and fate in the environment, as well as ecotoxicological effects are not entirely known. Therefore, it is required to include them in existing regulations focused on the environmental quality.
In this paper, a detail analysis of regulations dealing with the aquatic environment pollution, which represent a base for sustainable water resource management, was performed. The EU legislation including Water Framework Directive, “watch list” of pollutants and REACH Directive, were reviewed. Additionally, Guidelines for water quality monitoring and protection of aquatic organisms by USA Environmental Protection Agency were analyzed, as well as ISO 14000, a series of environmental management standards and ISO 24510, 24511 and 24512 standards for activities relating to drinking water supply systems and wastewater sewage systems. Concept of emerging pollutants management in aquatic environment is also presented.

AB  - Photocatalytic degradation of the methomyl pesticide in aqueous suspension
of TiO2 using an Osram ultra-vitalux® lamp (315-400 nm) was investigated.
The effect of parameters such as initial concentration of catalyst, initial
methomyl concentration, and pH were studied. Complete removal of
methomyl was achieved but mineralization of organic carbon was
incomplete indicating the formation of methomyl degradation products.
Liquid chromatography tandem mass spectrometry technique was used in
order to identify the methomyl degradation products and three major
degradation products have been identified.

AB  - U cilju poboljšanja kvaliteta leka i unapređenja sigurnosti njegovog korišćenja, potrebno je praćenje stabilnosti i određivanje degradacionih proizvoda koji mogu nastati tokom proizvodnje, rukovanja ili čuvanja leka. Cilj ovog rada je ispitivanje stabilnosti i praćenje degradacije višefun-kcionalnog leka anizomicina. Stabilnost anizomicina je ispitivana sprovođenjem studija forsirane degradacije, koje obuhvataju ispitivanje osetljivosti leka na hidrolizu (kiselu, baznu i neutralnu), oksidaciju, svetlost (vidljivu i UV) i povišenu temperaturu, u skladu sa ICH (eng. International Conference on Harmonization) smernicama. Za praćenje degradacije leka i identifikaciju degradacionih prozvoda korišćena je metoda tečne hromatografije sa tandem masenom spek-trometrijom. Pokazano je da je anizomicin stabilan u prisustvu oksidacionog sredstva i svetlosti, umereno stabilan u kiseloj i neutralnoj sredini, jako nestabilan u baznoj sredini i termo-degradabilan. Deacetilanizomicin je identifikovan kao glavni proizvod degradacije anisomicina.
AB  - In order to improve the quality of a drug and its safety usage, it is necessary to monitor stability
of a drug and to determine degradation products that may form during production, usage and
storage. Aim of this work was to investigate stability and monitor degradation of the
multifunctional drug anisomycin. Stability of anisomycin was studied by performing force
degradation that includes investigation of susceptibility of the drug to hydrolysis (acidic, alkaline
and neutral), oxidation, light (visible and UV) and temperature, in accordance with ICH
(International Conference on Harmonization) guidelines. Liquid chromatography coupled to
tandem mass spectrometry was used for investigation of the drug degradation and identification
of the degradation products. It was shown that anisomycin was stable in the presence of
oxidizing agent and light, moderately stable under acidic and neutral conditions, very unstable
under alkaline conditions and thermodegradable. Deacetylanisomycin was identified as the
principal degradation product of anisomycin.

AB  - A selective, sensitive and fast method for extraction, identification and quantification of multifunctional drug anisomycin in various tissues and serum, based on liquid chromatography-tandem mass spectrometry, was developed, optimized and validated. The method was validated according to the FDA guidelines and generally demonstrated good selectivity, accuracy, precision, and stability of the analyte. In the optimized method, high extraction efficiency was achieved for tested tissues (heart, brain, spleen, kidney, liver and femoral muscle) and serum. The obtained values of lower limits of quantification (LLOQ, 1.0-11.0 ng g(-1)) and limits of detection (0.3-3.3 ng g(-1)) indicated that the method was suitable for determination of trace levels of anisomycin in the complex matrices. The method was linear (R-2  gt = 0.990) in the tested concentration range (LLOQ-2500 ng g-1). The developed method was successfully applied in the first study on in vivo pharmacokinetics and distribution of anisomycin in tissues and serum of Wistar albino rats following subcutaneous injection (150 mg kg(-1)). The peak concentration in most tissues was achieved within 3 h after injection. The highest anisomycin concentration was found in the brain and the lowest concentration was found in the serum.

AB  - Predmet ovog rada bio je ispitivanje mogućnosti primene višeslojnih ugljeničnih nanocevi kao sorbenta u metodi ekstrakcije na čvrstoj fazi (eng. solid phase extraction SPE) pri analizi ostataka lekova iz vode. U cilju dobijanja SPE sorbenata različitih karakteristika, izvršeno je hemijsko modifikovanje ugljeničnih nanocevi korišćenjem 4 M KOH, 5 M HNO3 i 1 M HCl. Lekovi odabrani za analizu (metoprolol, sulfametaksazol, azitromicin, bisoprolol, enalapril, eritromicin, karbamazepin, lorazepam, klopidogrel, diklofenak, simvastatin i cilazapril) pripadaju grupama antibiotika, kardiovaskularnih lekova, analgoantipiretika i lekova za lečenje nervnog sistema, a to su istovremeneno najčešće korišćeni lekovi u Republici Srbiji. Pored lekova odabrana su i dva metabolita metamizola (4-AAA i 4-FAA), koji su u prethodnim istraživanjima često detektovani u uzorcima površinskih i podzemnih voda [2]. Za identifikaciju i kvantitativno određivanje analita korišćena je tečna hromatografija visoke performanse u sprezi sa tandem masenom spektrometrijom.

AB  - Lekovi predstavljaju najznačajniju grupu novih zagađujućih supstanci koje se nalaze u prirodnim vodama [1]. Otpadne vode, poreklom iz domaćinstava, industrije i životinjskih farmi predstavljaju osnovne izvore zagađenja životne sredine. U prirodnim vodama, lekovi mogu biti prisutni u svojoj osnovnoj formi ili u formi metabolita. Koncentracije metabolita lekova koje su prisutne u životnoj sredini često su veće od koncentracija osnovnog leka. Metamizol je veoma popularan antipiretik i analgetik koji se koristi u mnogim zemljama preko 70 godina. Iako je, zbog sumnje da može da izazove agranulocitozu kod pacijenata, zabranjen u mnogim zemljama, u većini evropskih zemalja i dalje se koristi zbog svog analgetičkog dejstva. Posle apsorpcije metamizola u gastro-intestinalnom traktu, on potpuno hidrolizuje u 4-metil-aminoantipirin (4-MAA), koji u jetri metabolizuje u 4-aminoantipirin (4-AA), pa se dalje acetilizuje do 4-acetilaminoantipirina (4-AAA) i 4-formil-aminoantipirina (4-FAA) [2]. Krajnji metaboliti 4-AAA i 4-FAA su dosta stabilni u životnoj sredini i nađeni su u dosta visokim koncentracijama u otpadnim i prirodnim vodama [3]. U ovom radu je optimizovana, validirana i primenjena analitička metoda za određivanje metamizola i njegovih metabolita 4-AAA i 4-FAA u podzemnim i površinskim vodama.

abstract = "Lekovi predstavljaju najznačajniju grupu novih zagađujućih supstanci koje se nalaze u prirodnim vodama [1]. Otpadne vode, poreklom iz domaćinstava, industrije i životinjskih farmi predstavljaju osnovne izvore zagađenja životne sredine. U prirodnim vodama, lekovi mogu biti prisutni u svojoj osnovnoj formi ili u formi metabolita. Koncentracije metabolita lekova koje su prisutne u životnoj sredini često su veće od koncentracija osnovnog leka. Metamizol je veoma popularan antipiretik i analgetik koji se koristi u mnogim zemljama preko 70 godina. Iako je, zbog sumnje da može da izazove agranulocitozu kod pacijenata, zabranjen u mnogim zemljama, u većini evropskih zemalja i dalje se koristi zbog svog analgetičkog dejstva. Posle apsorpcije metamizola u gastro-intestinalnom traktu, on potpuno hidrolizuje u 4-metil-aminoantipirin (4-MAA), koji u jetri metabolizuje u 4-aminoantipirin (4-AA), pa se dalje acetilizuje do 4-acetilaminoantipirina (4-AAA) i 4-formil-aminoantipirina (4-FAA) [2]. Krajnji metaboliti 4-AAA i 4-FAA su dosta stabilni u životnoj sredini i nađeni su u dosta visokim koncentracijama u otpadnim i prirodnim vodama [3]. U ovom radu je optimizovana, validirana i primenjena analitička metoda za određivanje metamizola i njegovih metabolita 4-AAA i 4-FAA u podzemnim i površinskim vodama.",

AB  - Zagađujuće materije u životnoj sredini, poput lekova i pesticida ispoljavaju veliki afinitet da se akumuliraju u rečnim sedimentima, tako da rečni sedimenti mogu postati sekundarni izvor zagađenja. AB  - Cilj ovog rada bio je ispitivanje načina adsorpcije i raspodele odabranih lekova i pesticida između vodene faze i sedimenta, kao i određivanje stepena adsorpcije i desorpcije na sedimentu reke Dunav. Rezultati adsorpcije leka karbamazepina i pesticida atrazina, karbendazima i karbofurana pokazuju najbolje slaganje sa Frojndlihovom izotermom, što ukazuje na reverzibilnu adsorpciju. Zbog visokog procenta desorpcije karbamazepina, atrazina i karbendazima, njihovo prisustvo u sistemu voda-sediment je očekivano, dok se karbofuran očekuje u manjoj koncentraciji u vodi, zbog nižeg procenta desorpcije. Lengmirov model opisuje monoslojnu adsorpciju pesticida dimetoata. Ovaj pesticid pokazuje i mali stepen desorpcije, što ukazuje na to da se može akumulirati u sedimentima u visokoj koncentraciji. Adsorpcija lekova trimetoprima, metoprolola i lorazepama se može opisati linearnim modelom. Ovi analiti pokazuju veliki afinitet adsorpcije i mogu se vezati za sediment u koncentracijama većim od ispitivanih. Za razliku od metoprolola i lorazepama, trimetoprim se u velikom procentu desorbuje sa sedimenta, pa se ovaj analit očekuje u vodi. Lekovi bromazepam, eritromicin i amlodipin su pokazali skoro potpunu adsorpciju na sedimentu i veoma mali stepen desorpcije, pa je njihova akumulacija u sedimentima očekivana.
AB  - The aim of this study was investigation of the adsorption and distribution of the selected
pharmaceuticals and pesticides between water phase and sediment, as well as determination
of the adsorption and desorption extent using sediment of the Danube River. Adsorption
data of the drug carbamazepine and pesticides atrazine, carbendazim and carbofuran fit well
to Freundlich isotherm, implying reversible adsorption. Due to high desorption extent of
carbamazepine, atrazine and carbendazim, their presence in water-sediment system is
expected, whereas lower desorption level of carbofuran indicates lower expected
concentration of this pesticide in water. Langmuir model describes monolayer adsorption of
pesticide dimethoate. This pesticide also shows low desorption extent, indicating its high
accumulation in sediments. Adsorption data of pharmaceuticals trimethoprim, metoprolol
and lorazepam fit fine to the linear model. These analytes show high adsorption affinity and
can be adsorbed onto sediment at concentrations higher than tested. Unlike metoprolol and
lorazepam, trimethoprim exhibits higher desorption level, so it is expected in water.

adsorption onto sediment and extremely low desorption rate, so their accumulation in

AB  - Anisomycin is an immunosuppressant in low doses ( lt 0.1 mu M) with possible application in treatment of some autoimmune diseases and in inhibiting transplantation rejection. Anisomycin suppresses malignant tumor cell growth and affects memory. For the first time it was the subject of the electrochemical investigations by cyclic voltammetry and square wave voltammetry on gold electrode in 0.05 M NaHCO3 using its electrochemical activity. The cyclic voltammetry experiments at different sweep rates show that electrochemical process is irreversible and diffusion controlled. Based on square wave voltammetry measurements, the calculated values of LOD and LOQ were 1 and 4 nM (in the absence of biological fluid), as well as 2 and 6 nM (in the presence of spiked urine) indicating the high sensitivity of the proposed electroanalytical method. High performance liquid chromatography-tandem mass spectrometry was a reference method for quantification of anisomycin and served for structural identification of its hydrolysis product (deacetylanisomycin).

Determination of antibiotic anisomycin in tissue samples by liquid chromatography–tandem mass spectrometry

AB  - The aim of this work was development of sensitive analytical method for determination of drug anisomycin in different tissue samples. The optimized extraction procedure included methanol as extraction solvent and Oasis HLB as extract clean up cartridge. In the method validation, ani-somycin was efficiently extracted from most of tissues, such as heart, brain, spleen, kidney and li-ver, whereas lower recoveries were obtained for fat tissue. Low limits of detection (0.6–3.8 ng g–1) and quantification (2.4–11.8 ng g–1) indicate that developed method is suitable for determination of anisomycin trace levels. The method was linear (R2 > 0.975) in the tested concentration range (50–2500 ng g–1). Significant matrix effect was eliminated using matrix-matched standards. It was finally determined that the developed method is sensitive and reliable for determination of drug anisomycin in various tissue samples using liquid chromatography–tandem mass spectrometry.
AB  - Cilj ovog rada bio je razvoj osetljive analitičke metode za određivanje leka anizomicina u različitim tkivima. U
optimizovanoj proceduri ekstrakcije, metanol je odabran kao rastvarač za ekstrakciju, a Oasis HLB kao
kertridž za prečišćavanje ekstrakta. Prilikom validacije razvijene metode, anizomicin je efikasno ekstrahovan
iz većine tkiva, kao što su srce, mozak, slezina, bubrezi i jetra, dok su za masno tkivo dobijeni niži prinosi.
Niske granice detekcije (0,6–3,8 ng g
–1
) i kvantifikacije (2,4–11,8 ng g–1
) ukazuju da je razvijena metoda
pogodna za određivanje tragova anizomicina. U ispitivanom opsegu koncentracija (50–2500 ng g–1
) metoda je
linearna (R2
> 0,975). Značajan uticaj matrice eliminisan je upotrebom standarda koji odgovaraju matrici
uzorka. Na osnovu rezultata validacije utvrđeno je da je razvijena metoda osetljiva i pouzdana za određivanje
leka anizomicina u različitim tkivima korišćenjem tečne hromatografije sa tandem masenom spektrometrijom.

It was finally determined that the developed method is sensitive and reliable for determination of drug anisomycin in various tissue samples using liquid chromatography–tandem mass spectrometry., Cilj ovog rada bio je razvoj osetljive analitičke metode za određivanje leka anizomicina u različitim tkivima. U
optimizovanoj proceduri ekstrakcije, metanol je odabran kao rastvarač za ekstrakciju, a Oasis HLB kao
kertridž za prečišćavanje ekstrakta. Prilikom validacije razvijene metode, anizomicin je efikasno ekstrahovan
iz većine tkiva, kao što su srce, mozak, slezina, bubrezi i jetra, dok su za masno tkivo dobijeni niži prinosi.
Niske granice detekcije (0,6–3,8 ng g
–1
) i kvantifikacije (2,4–11,8 ng g–1
) ukazuju da je razvijena metoda
pogodna za određivanje tragova anizomicina. U ispitivanom opsegu koncentracija (50–2500 ng g–1
) metoda je
linearna (R2
> 0,975). Značajan uticaj matrice eliminisan je upotrebom standarda koji odgovaraju matrici
uzorka. Na osnovu rezultata validacije utvrđeno je da je razvijena metoda osetljiva i pouzdana za određivanje
leka anizomicina u različitim tkivima korišćenjem tečne hromatografije sa tandem masenom spektrometrijom.",

AB  - Anizomicin je antibiotik izolovan iz bakterijske vrste Streptomyces griseolus za koji je nekoliko studija pokazalo da je citotoksičan prema ćelijama malignog tu-mora, tj. da može suzbiti njihov rast. Cilj ovog rada bio je razvoj i optimizacija metode  ekstrakcije  ovog  antibiotika  iz  životinjskih  tkiva,  kao  što  su  srce  i  jetra.  Ispitivani  su  različiti  rastvarači  za  ekstrakciju,  kao  i  metode  prečišćavanja  dobijenog ekstrakta. Ekstrakt je analiziran metodom tečne hromatografije sa tandem masenom  spektrometrijom  (LC–MS/MS,  engl.  liquid  chromatography–tandem  mass spectrometry).
