AB  - Selenium (Se) is an essential trace element for human health, but its nutritional supply is insufficient in large parts of the world. Mushrooms can be enriched in selenium and can serve as alternative and natural source of selenium supplementation. In the present study, two common mushroom species (Pleurotus ostreatus and Ganoderma lucidum), were enriched with two selenium compounds (selenite and selenate) to test their suitability as natural sources of selenium supplementation. Sharp differences in the the metabolic patterns of the fortified selenium were observed. Selenium was effectively metabolized in P. ostreatus but remained in inorganic form in G. lucidum. However, mushrooms extracts were effective in enhancing selenoprotein expression in cell lines. The present study highlights the importance of employing selenium speciation analysis with an element-selective technique to examine the metabolic products following mushroom fortification for nutritional purposes due to the different toxicological profile and bioavailability of different selenium biotransformation products.

AB  - Background This study provides an insight into the impact of ultrasound-assisted extraction with water as solvent (UAEW) and extraction by supercritical carbon dioxide (SC-CO2) with 5% EtOH on antioxidant and enzyme inhibitory activity in regard to the chemical profile of the edible and medicinal mushroom,Pleurotus pulmonarius. Results Extraction efficiency was between 0.36% and 63.32%, depending on the extraction technique. The main compounds in the extracts were fatty acids. Supercritical CO(2)extraction with co-solvent was the most suitable method for obtaining extracts that were rich in ergosterol content, reaching a value of 40.1 mg g(-1). The UAEW of crude mushroom powder ensured the highest yield, as well as the extracts with best antioxidative activity. The measurements of enzyme inhibitory activity revealed that all types of investigated extracts exhibited only tyrosinase and amylase inhibition at a significant level. Conclusion Based on our results, the extraction methods significantly affected the chemical profile and bioactivity ofP. pulmonarius.

Simultaneous selenium and sulfur speciation analysis in cultivated Pleurotus pulmonarius mushroom

Mycelium, colonized substrate, and fruiting bodies at different harvesting times were analyzed using ion-pairing reversed-phase chromatography coupled to an ICPMS/MS detector. Extraction efficiencies in en-zymatically digested and aqueous extracts were between 45.3 and 109% for Se, depending on the sample type. Selenomethionine (Se-Met) was found to be the major Se-compound, together with a number of unknown Se-species. Cystine (Cys(2)), methionine (Met), and sulfate were also detected and quantified in all samples. Most of the Se-Met (84.0%) and met (75.8%) were found to be in free form in the fruiting body, in contrast with the mycelium where 53.4% of Se-Met and 80.5% of met is incorporated into proteins.

Wheat Straw Degradation by Trametes gibbosa: The Effect of Calcium Ions

AB  - Wheat straw is the major crop residue in Europe which makes it the most promising material for bioconversion. However, only 3% of annual production is used directly while the rest is considered as waste. A key step in processing of lignocellulosics is delignification by fungi and the presence of calcium can be of a great importance as it can modulate ligninolytic enzymes activities. Trametes gibbosa BEOFB 310 was used for solid-state fermentation of wheat straw in the presence of calcium in different concentrations. Samples were extracted after 19days of fermentation and activities of Mn-oxidizing peroxidases and laccase were determined spectrophotometrically. Quantitative procedures were used to determine contents of hemicelluloses, cellulose, and lignin. Calcium induced activity of Mn-dependent peroxidase to a concentration of 5.0mM (7185.2 +/- 791.4 UL-1), which was threefold higher than in the control. Lignocellulose loss in wheat straw was stimulated by calcium addition and the maximum delignification was detected at concentration of 5.0mM (52.9 +/- 0.9%). Delignification was positively correlated to activity of Mn-dependent peroxidase. This study showed that wheat straw supplementation with calcium can significantly improve solid-state fermentation by increasing Trametes gibbosa Mn-dependent peroxidase activity and augmenting lignin degradation.

Antioxidative, antifungal, cytotoxic and antineurodegenerative activity of selected Trametes species from Serbia

AB  - In a last few decades mushrooms are increasingly attracting attention as functional food and sources of biologically active compounds. Several Trametes species have been used for centuries in traditional medicine of East Asia cultures, but only T. versicolorwas studied sufficiently while there are less substantial data about medicinal properties of other species. Trametes versicolor, T. hirsuta and T. gibbosa were the species tested for biological activities. Antifungal potentials of extracts were assessed for clinical strains of selected Candida and Aspergillus species. ABTS and FRAP assays were used to evaluate antioxidant capacities of studied extracts. Cytotoxic activity was determined against human cervix and lung adenocarcinoma and colon carcinoma cell lines. Antineurodegenerative activity was assessed by determining the rate of acetylcholinesterase and tyrosinase activity. The presence of metabolites in extracts of mycelia and basidiocarps of studied Trametes species was analyzed by H-1 NMR spectroscopy. Studied extracts showed low antifungal potential in comparison with ketoconazole. Basidiocarp extracts were more effective ABTS(+) scavengers and Fe2+ reducers than mycelium ones but less effective in comparison with L-ascorbic acid. Results showed that mycelium extracts had stronger cytotoxic effects against three cancer cell lines than basidiocarp ones, and that cervix adenocarcinoma cells were the most sensitive to the extracts and commercial cytostatics. T. versicolor mycelium extract was the most effective inhibitor of acetylcholinesterase activity but double weaker than galantamine, and T. gibbosa mycelium extract was significantly better inhibitor of tyrosinase activity than kojic acid for 40.9%. Chemical analysis indicated strong synergistic action of triterpenes, sugars and polyphenols in applied assays. The results suggest that tested Trametes species have significant medicinal potentials which could be attributed to antioxidative and cytotoxic activity. Additionally both, basidiocarps and mycelia extracts can strongly inhibit activity of acetylcholinesterase and tyrosinase.

Degradation of beech wood and wheat straw by Trametes gibbosa

AB  - The aim of this study was to optimize cultivation conditions for Mn-oxidizing peroxidases and laccase production and selective degradation of beech wood and wheat straw lignin by Trametes gibbosa. To promote lignin degradation, the effect of different carbon and nitrogen sources, type of cultivation, enzyme dynamics and inducers were studied. Solid-state cultivation was optimal for lignin degradation. Wheat straw was the optimal carbon source for peroxidase activities stimulating the synthesis of numerous isoforms. The dynamics of enzymatic activities showed that 19-day-old fermentation is optimal and that activities were directly associated with enzyme production. Alteration of nitrogen sources and inducers was applied to increase the rate of lignin degradation and decrease cellulose degradation. The presence of nitrogen in the form of (NH4)(2)SO4 and concentration of 10 mM significantly increased the extent and selectivity of delignification of wheat straw compared with cellulose (44.1 vs. 36.1%). The most effective and selective wheat straw degradation (52.0% of lignin vs. 31.3% of cellulose) was achieved by enriching the optimum medium, as defined above, with 1.0 mM p-anisidine. The optimum conditions for wheat straw processing to achieve delignification and production of highly active ligninolytic enzymes by T. gibbosa were (NH4)(2)SO4/wheat bran medium supplemented with p-anisidine. This study indicated the significant potential of optimizing external factors as a promising tool for induction and selectivity in the degradation of lignocelluloses by T. gibbosa as a pretreatment for several biotechnological processes.

AB  - Due to a high content of rutin (2-10%), dried buckwheat leaf and flower (DBLF) formulations were shown to be efficient in the treatment of vascular diseases. In order to find a cost effective way for the extraction of antioxidants, the effects of ethanol/water ratio and temperature on the extraction efficiency of phenolic compounds and the mechanisms of antioxidant action of the extracts were tested. Extraction with ethanol/water mixture (80:20, v/v) for 24 h at room temperature, after the mixture was just brought to boil was demonstrated to be an efficient and cheap way for obtaining a high yield of rutin (49.94 +/- 0.623 mg/g DBLF). The most abundant phenolic compounds in DBLF extracts were rutin and chlorogenic acid. Flavonoids, especially rutin, were shown to be the most responsible for the antioxidant activity in all investigated lipid model systems, acting as free radical scavengers, electron-donating substances and chelators of iron ions. In beta-carotene bleaching tests, the extracts with the highest activity were as efficient as BHT (butylated hydroxytoluole). Regarding the results of antihemolytic and Schaal oven tests, the extracts demonstrated remarkable ability to inhibit the oxidative destruction of erythrocytes and to prolong the beginning of the oxidation process in sunflower oil.

AB  - Wheat straw is the major crop residue in European countries which makes it the most promising material for bioconversion into biofuels. However, cellulose and hemicellulose are protected with lignin, so delignification is an inevitable phase in lignocellulose processing. The organisms predominantly responsible for its degradation are white-rot fungi and among them Trametes species represent promising degraders due to a well-developed ligninolytic enzyme system. Although numerous studies have confirmed that low molecular weight compounds can induce the production and activity of ligninolytic enzymes it is not clear how this reflects on the extent of delignification. The aim of the study was to assess the capacity of p-anisidine and veratryl alcohol to induce the production and activity of Mn-oxidizing peroxidases and laccases, and wheat straw delignification by six Trametes species. Significant inter- and intraspecific variations in activity and features of these enzymes were found, as well as differences in the potential of lignocellulose degradation in the presence or absence of inducers. Differences in the catalytic properties of synthesized enzyme isoforms strongly affected lignin degradation. Apart from enhanced lignin degradation, the addition of p-anisidine could significantly improve the selectivity of wheat straw ligninolysis, which was especially evident for T. hirsuta strains.

AB  - The goal of this study was the evaluation of antifungal, antioxidant and anticancer potentials of Pleurotus eryngii, P. ostreatus and P. pulmonarius mycelial extracts, and the influence of mycelium enrichment with selenium on these activities. Both Se-amended and non-amended extracts showed the same or similar minimal inhibitory concentration for 14 studied micromycetes, while a fungicidal effect was not noted, contrary to ketoconazole, which had inhibitory and fungicidal effects at very low concentrations. Se-non-amended extracts exhibited antioxidant activity, especially at higher concentrations. Selenium enrichment influenced activity, its effects decreasing in P. eryngii and P. pulmonarius, while in P. ostreatus no effect was noted. The DPPH radical scavenging capacity of the extracts was in direct correlation with their phenol and flavonoid contents. Cytotoxic activity against both HeLa and LS174 cell lines was very low compared with cis-DDP. These features suggest that mycelium should be an object of intensive studies.

AB  - The aim of this study was to evaluate the effect of high selenium (Se) concentrations on morphophysiological and ultrastructural properties of Pleurotus ostreatus. Mycelium growth was good in media enriched with 5.0, 10.0, and 20.0 mg L-1 of Se, concentration of 500.0 mg L-1 strongly inhibited growth, and 1000.0 mg L-1 was the minimum inhibitory concentration. Contrary to thin-walled, hyaline, branched, and anastomized hyphae with clamp-connections in the control, at Se concentrations of 100.0 and 500.0 mg L-1, they were noticeably short, frequently septed and branched, with a more intensive extracellular matrix, and without clamp-connections. At high Se concentrations, hyphae with intact membrane, without cellular contents, with a high level of vacuolization, and with numerous proteinaceous bodies were observed. Biomass yield ranged between 11.8 g L-1, in the control, and 6.8 g L-1, at an Se concentration of 100.0 mg L-1, while no production was detected at a concentration of 500.0 mg L-1. Se content in the mycelia reached a peak (938.9 mu g g(-1)) after cultivation in the medium enriched with Se at the concentration of 20.0 mg L-1, while the highest absorption level (53.25%) was found in the medium enriched with 5.0 mg L-1 Se.

AB  - The aim of the study was comparative analysis of ligninolytic enzyme productions of Trametes gibbosa and its ability to degrade wheat straw, as well as effect of trace elements on those processes. Mn-oxidizing peroxidases were the main enzymes and the tested trace elements affected their activities mainly inhibitory. Stimulatory effect on Mn-independent peroxidase had Mn2+ in all concentrations, while Zn2+ concentrations of 5.0 rnM and 10.0 mM led to the maximum Mn-dependent peroxidase activity. The trace elements affected laccase activity differently. Zn2+ and Fe2+ (0.5 mM and 10.0 mM, respectively) had the greatest effect on the activity, while Mn2+ and Cu2+ at the amount of 10.0 mM completely stopped the enzyme production. The tested enzymes acted simultaneously on wheat straw degradation. The presence of 1.0 mM of Mn2+ caused the highest level of dry matter loss (43.1%), which was the consequence of hemicellulose, cellulose and lignin degradation in the rate of 62.6%, 35.6% and 61.2%, respectively. Selective lignin degradation was noted especially in the presence of Fe2+ at the concentration of 0.5 mM.

Zn2+ and Fe2+ (0.5 mM and 10.0 mM, respectively) had the greatest effect on the activity, while Mn2+ and Cu2+ at the amount of 10.0 mM completely stopped the enzyme production. The tested enzymes acted simultaneously on wheat straw degradation. The presence of 1.0 mM of Mn2+ caused the highest level of dry matter loss (43.1%), which was the consequence of hemicellulose, cellulose and lignin degradation in the rate of 62.6%, 35.6% and 61.2%, respectively. Selective lignin degradation was noted especially in the presence of Fe2+ at the concentration of 0.5 mM.",

Potential of Trametes species to degrade lignin

AB  - Well developed ligninolytic enzyme system in white-rot mushrooms is responsible for degradation of low degradable and lignin rich unused plant debris, which represents serious environmental ballast. The biological degradation is environmentally friendly and economically justified process. Therefore, assessment of the potential of Trametes pubescens and Trametes multicolor to degrade lignin of wheat straw and oak sawdust during solid-state cultivation was the object of the study. Significantly high laccase activity was noted on day 7 and day 10 of wheat straw fermentation by T pubescens BEOFB 330, while at other days the obtained values were lower from 2- to 8-fold. Contrary to laccase, the optimum substrate for Mn-oxidizing peroxidases was oak sawdust. Maxima activities of Mn-dependent peroxidase and Mn-independent peroxidase were noted on day 14 of cultivation of T. multicolor HAI 426 and T. pubescens BEOFB 330, respectively. Tested organisms, except T multicolor HAI 428, have degraded about 50% of lignin in wheat straw that was more degradable residue than oak sawdust, which decay level ranged between 1.8% and 5.9%. Mn-oxidizing peroxidases had the main role in oak sawdust lignin degradation. T. multicolor HAI 426, was the best lignin degrader in which laccase activity was not noted.

AB  - Global industrialization and agricultural development are related to the release of various pollutants into the environment including huge amounts of free radicals, which are associated with the occurrence of various destructive diseases and disorders. Due to different rates of metabolic activity and oxygen consumption, organisms, organs and tissues have distinct protective antioxidant systems and defence mechanisms. Although numerous synthetic antioxidants can improve defence system capacity, because of their toxic and mutagenic effects as well as rising awareness of a healthy lifestyle, preference is given to natural antioxidants. Therefore, studies of metabolites of various fungal species and their activity are currently increasing. Antioxidant features of numerous compounds isolated from mushrooms, such as phenolic compounds, vitamins, polysaccharides, peptides, proteins, organic acids, carotenoids, alkaloids, and nucleotides, have been reported. Chemical composition and antioxidant potential of mushrooms are highly-dependent on species, habitat, phase of life cycle (mycelium, young or mature fruiting body), method of processing, extraction solvent, and extract dose. Although around only 5% of fungal species have been well studied and thousands more species have potential benefit for mankind, reports on the antioxidant potentials of fungi are already numerous and these are the subject of this review.

AB  - Trace elements, at tolerable concentrations, are either part of the active site or act as an activity modulator of ligninolytic enzymes of white-rot mushrooms. They are usually in plant raw materials in a non-toxic amount or non-available and extractable forms. This study evaluated the effects of Fe, Zn, and Se on the activity of laccase and Mn-oxidizing peroxidases of P. ostreatus and P. pulmonarius during solid-state fermentation of grapevine sawdust. The studied species showed different levels of tolerance to the trace elements. A stimulatory effect of the microelements on laccase activity was demonstrated in P. ostreatus, while Fe and Zn were strong inhibitors of the activity of P. pulmonarius, which was contrary to Se, independent of the cultivation period. With the exception of SeO2, activity of Mn-oxidizing peroxidases in P. ostreatus was suppressed in various levels by the elements. However, in P. pulmonarius, activity against phenol red oxidation in the presence of external Mn2+ was stimulated by the elements on day 7, while on day 10, activity inhibition by Fe and Zn and stimulation by Se was noted. The effect on activity against phenol red oxidation in the absence of external Mn2+ was the opposite.

AB  - The purpose of this study was to resolve the question of whether various selenium concentrations affect the ability of eight species, members of five genera, to produce mycelium biomass and absorb this trace element during submerged cultivation in Na2SeO3-enriched medium. The obtained results demonstrated the ability of mycelia of all of the tested species to absorb selenium at analyzed concentrations. Thus, selenium content ranged from 1.4 mu g g(-1) (Pleurotus eryngii) to 20.3 mu g g(-1) (Pleurotus ostreatus). The potential of mycelia to absorb selenium was significant, ranging from 8.1% (Lenzites betulinus) to 62.5% (P ostreatus) of its content in the medium. It may be concluded that all tested mushroom species could be used as satisfactory selenium sources due to the fact that the absorbed concentrations ranged from 15.8% (P eryngii) to 36.9% (P. ostreatus) of the dietary selenium amount (55 mu g d(-1)) recommended by the European Scientific Committee on Food.

AB  - As biological decomposition of plant biomass represents a popular alternative environmental-friendly and economically justified process, screening of ligninolytic enzyme systems of various fungal species is a topical study area. The goal of the study was to obtain clear insight into the dynamics of laccase, Mn-dependent peroxidase, and Mn-independent peroxidase activity and levels of wheat straw lignin degradation in seven wood-rotting fungi. The best laccase producers were Pleurotus ostreatus and Pleurotus eryngii. Lenzites betulinus and Fomitopsis pinicola were the best Mn-dependent peroxidase producers, and P. ostreatus the weakest one. The peak of Mn-independent peroxidase was noted in Dichomytus squalens, and the minimum value in P. ostreatus. The profiles of the three enzymes, obtained by isoelectric focusing, were variable depending on the species and cultivation period. D. squalens was the best lignin degrader (34.1% of total lignin amount), and P. ostreatus and P. eryngii the weakest ones (7.1% and 14.5%, respectively).
