Biocompatibility and antimicrobial activity of zinc(II)-doped hydroxyapatite, synthesized by a hydrothermal method

AB  - In order to obtain multifunctional materials with good biocompatibility and antimicrobial effect, hydroxyapatite (HAp) doped with Zn2+ was synthesized by a hydrothermal method. Powders with different content of zinc ions were synthesized and compared with undoped HAp to investigate the influence of Zn2+ on the antimicrobial activity of HAp. Analyses of undoped and Zn2+-doped powders before and after thermal treatment at 1200°C were performed by SEM and XRD. The antimicrobial effects of the powders were examined in relation to Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Candida albicans in liquid medium. The results showed that the obtained powders had good antimicrobial activity, but higher antimicrobial activities of the powders doped with Zn2+ were observed after annealing at 1200°C. For powders annealed at 1200°C, in vitro biocompatibility tests: reduction of the tetrazolium salt 3-(4,5-dimethylthazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT test) and Trypan Blue dye exclusion test (DET test) with MRC-5 fibroblast cells in liquid medium were performed. Based on the MTT and DET tests, it was shown that the powders do not have a significant cytotoxic effect, which was confirmed by SEM analysis of MRC-5 fibroblast cells after their in vitro contact with the powders.
AB  - U cilju dobijanja multifunkcionalnih materijala sa dobrom biokompatibilnošću i dobrim antimikrobnim dejstvom, hidroksiapatit (HAp) sa ugrađenim Zn2+ sintetisan je hidrotermalnom metodom. Za potrebe istraživanja uticaja Zn2+ na anti-mikrobnu aktivnost, sintetisani su HAp prahovi sa različitim sadržajem Zn2+ i upoređeni sa čistim HAp. Čisti prahovi i oni sa ugrađenim cinkom, pre i posle termičkog tretmana na 1200°C, analizirani su skenirajućom elektronskom mikroskopijom (SEM) i metodom rendgenske difrakcije. Antimikrobni efekat prahova ispitivan je prema sojevima Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa i Candida albicans u tečnom medijumu. Antimikrobni test je pokazao da dobijeni prahovi imaju dobru antimikrobnu aktivnost. Poboljšana antimikrobna aktivnost prahova u koje je ugrađen Zn2+ postignuta je žarenjem na 1200°C. In vitro testovi biokompatibilnosti, MTT i DET, sa MRC-5 fibroblastnim ćelijama u tečnom medijumu urađeni su za prahove žarene na 1200°C. Na osnovu MTT i DET testova zaključeno je da prahovi nemaju značajan citotoksični uticaj što je potvrđeno SEM analizom MRC-5 fibroblastnih ćelija nakon njihovog in vitro kontakta sa prahovima.

AB  - The effect of decreasing the grain size on the biocompatibility, cellmaterial interface, and mechanical properties of microwave-sintered monophase hydroxyapatite bioceramics was investigated in this study. A nanosized stoichiometric hydroxyapatite powder was isostatically pressed at high pressure and sintered in a microwave furnace in order to obtain fine grained dense bioceramics. The samples sintered at 1200 degrees C, with a density near the theoretical one, were composed of micron-sized grains, while the grain size decreased to 130 nm on decreasing the sintering temperature to 900 degrees C. This decrease in the grain size certainly led to increases in the fracture toughness by much as 54%. An in vitro investigation of biocompatibility with L929 and human MRC-5 fibroblast cells showed noncytotoxic effects for both types of bioceramics, while the relative cell proliferation rate, cell attachment and metabolic activity of the fibroblasts were improved with decreasing of grain size. An initial in vivo investigation of biocompatibility by the primary cutaneous irritation test showed that both materials exhibited no irritation properties.

Multifractal analysis of the UV/VIS spectra of malignant ascites: Confirmation of the diagnostic validity of a clinically evaluated spectral analysis

AB  - Multifractal (MF) approach was applied for the analysis of ultraviolet/visible (UV/VIS) spectra as an independent confirmation of the diagnostic efficacy of UV/VIS spectral analysis of intraperitoneal fluids, ascites, taken from patients with a known clinical diagnosis. Recently, it was reported that from UV/VIS spectra differentiation of malignant from benign ascites is possible. Here, it was shown that by using MF analysis of UV/VIS spectra, the objective classification of UV/VIS spectra is possible. The applicability of UV/VIS analysis and MF classification of spectra were evaluated on N = 68 cases, of which M = 64 and B = 4 were clinically confirmed as malignant and benign, respectively. The overall diagnostic efficacy was 89.71% when using on-line analysis of UV/VIS spectra (61 out of 68 samples were positively recognized: 58 malignant and 3 benign), and even 95.59% by using off-line MF classsification (65 out of 68 samples were classified correctly: 63 malignant and 2 benign). It can be inferred that UV/VIS spectral analysis of ascites, combined with MF analysis, could be suggested as a successful and safe screening method in the evaluation of intraperitoneal fluids.

AB  - Atomic force microscopy (AFM), a unique tool to investigate drug treatment of cancer cells, was used to analyze the anti-neoplastic activity of adriamycin by comparing DNA structures of non-treated and adriamycin-treated Ehrlich tumor cells. The non-treated cells exhibited a highly branched intact chromatin structure, related to the intensive DNA replication in cancer cells. Images from adriamycin-treated tumor cells showed that the DNA chains were broken and the chromatin structure had been destroyed. Possible explanations for these effects of adriamycin are considered: breakage of hydrogen bonding, oxidation and intercalation effects, as well as the poisoning of topoisomerase enzyme. DNA fractal and multifractal analyses, performed in order to evaluate the degree of bond scission, showed that the treated DNA had become more fractal compared to non-treated DNA.
