Biomedical Applications of Thermosensitive Hydrogels for Controlled/Modulated Piroxicam Delivery

AB  - The objectives of this study are the synthesis of thermosensitive poly(N-isopropylacrylamide-co-2-hydroxypropyl methacrylate), p(NiPAm-HPMet), hydrogels and the analysis of a drug-delivery system based on piroxicam, as a model drug, and synthesized hydrogels. A high pressure liquid chromatography method has been used in order to determine both qualitative and quantitative amounts of unreacted monomers and crosslinkers from polymerized hydrogels. Swelling kinetics and the order of a swelling process of the hydrogels have been analyzed at 10 and 40 °C. The copolymers’ thermal properties have been monitored by the differential scanning calorimetry (DSC) method. DSC termograms have shown that melting occurs in two temperature intervals (142.36–150.72 °C and 153.14–156.49 °C). A matrix system with incorporated piroxicam has been analyzed by using FTIR and SEM methods. Structural analysis has demonstrated that intermolecular non-covalent interactions have been built between side-groups of copolymer and loaded piroxicam. Morphology of p(NiPAm-HPMet) after drug incorporation indicates the piroxicam presence into the copolymer pores. Kinetic parameters of the piroxicam release from hydrogels at 37 °C and pH 7.4 indicate that the fluid transport mechanism corresponds to Fickian diffusion. As a result, formulation of thermosensitive p(NiPAm-HPMet) hydrogels with incorporated piroxicam could be of interest for further testing as a drug carrier for modulated and prolonged release, especially for topical administration.

Temperature-Sensitive Hydrogels as Carriers for Modulated Delivery of Acetaminophen

AB  - The purposes of this study are the polymerization of temperature-sensitive copolymers based on N-isopropyl acrylamide and 10 mol % of 2-hydroxypropylmethacrylate, characterisations of their thermal, morphological and swelling properties, as well as the analysis of potential application in drug-delivery systems. Acetaminophen, the representative of non-steroidal anti-inflammatory drugs, was used as a model drug in this study. It is a common pain relief drug, which is also used for fever treatment. However, oral administration comes with certain health risks, mainly the overdose and frequent administration of up to four times a day. The goal of applying temperature-sensitive hydrogel is to enable extended administration once a day, depending on the body temperature. The swelling behavior of the obtained poly(N-isopropyl acrylamide-co-2-hydroxypropylmethacrylate) (p(NIPA/HPMA)) hydrogels and their temperature-sensitivity, kinetics and order of swelling processes at 18 and 38 °C were analyzed. The thermal properties of these hydrogels were observed by the DSC method, and the obtained thermograms showed both melting and glass transitions. The drug delivery system of p(NIPA/HPMA) hydrogels with loaded acetaminophen was analyzed using scanning electron microscopy and Fourier transform infrared spectroscopy methods. Structural analysis of FTIR spectra indicates that non-covalent intermolecular interactions of the type of hydrogen bonds were formed among functional groups of acetaminophen and side-chains of p(NIPA/HPMA) hydrogels. The surface structure of p(NIPA/HPMA) hydrogels after drug loading indicates the acetaminophen presence into the pores of the hydrogel network, and their loading efficiency was higher than 92%. Qualitative and quantitative analysis of acetaminophen, determined by the high-pressure liquid chromatography method, showed that about 90–99% of the loaded amount was released from p(NIPA/HPMA) hydrogels within 24 h. Kinetic parameters of the acetaminophen release under simulated gastrointestinal conditions were determined. Based on obtained results, the drug delivery system of temperature-sensitive p(NIPA/HPMA) hydrogels with loaded acetaminophen could be suitable for additional investigation for modulated drug administration, e.g., for extended drug administration.

AB  - Duloxetine hydrochloride (DUL) is effective in treating depression, and was examined for electroanalytical purposes. The DULstandard was investigated by cyclic voltammetry (CV) and determined using differential pulse voltammetry (DPV) via its electrooxidationat the Au electrode in 0.05 M NaHCO3. DPV showed a linear dependency of the anodic peak currents vs DUL standardconcentrations in the range from 0.1 to 3.33 μg ml−1 with the values of the limit of detection (LOD) and the limit of quantification(LOQ): 0.133 and 0.667 μg ml−1, respectively. Using the constructed and validated calibration curve, the values of unknown DULconcentrations in both Taita® tablets and in human serum spiked with the standard were determined. The number of protons andelectrons transferred was calculated and possible reaction mechanisms taking place on the surface of both electrodes wereproposed. The Au electrode exhibited a better sensitivity and a wider range of current vs concentration linear dependency for DULquantitative determination than the GC electrode. The study of DUL degradation showed that at the Au and GC electrodes, after4.5 h of potential cycling, degradation occurs, giving formaldehyde as a product, which was confirmed by high performance liquidchromatography (HPLC)

Computer–aided decision–assistant system for diagnosis of different diseases

AB  - The constant need to find an increasing number of diagnostic more selective techniques by which we would be able to determine an altered substrate as characteristic of the disease suspected is everyday clinical problem. It is important to note that diagnostic failure is the main cause of unnecessary costs that account for about 35% funds intended for healing in funds. The greatest number of people will experience at least one diagnostic error during their lifetime. The already realized results concerning new invented UV-vis method and automatic classification of the obtained spectroscopic signals in diagnostics of different diseases led to the the invention and construction of Computer-Aided Decision-Assistant system for Diagnosis of Different Diseases. It was achieved by automatization of UV-vis signals of different body fluids without or with known official diagnosis. In the invention and construction of this system is included clinical work, the softwer and hardwer realization, design and logo. Results included into the realization of constructed prototype will be presented.

Synthesis and FT-IR Study of Some N-Monosubstituted 2,2-Dimethylpropanamides

AB  - In our study of behaviour of the amide bond we investigated the conformations of some N-monasubstituted 2,2-dimethy|propanamides of the general formula CH3C(CH3)2CONHR, wherein R ranges from n-alkyl (C1-C7), iso-alkyil (C3-C6), sec-butyl, allyl, cycloalkyl (C3-C6), phenyl, to 4-substituted phenyl (where the substituents are fluoro, methyl, cyano, bromo, nitro, carboxymethyl, N,N-dimethylamino, methoxy, trifluoromethyl), 2-phenylethyl, and acetophenyl. The amides were synthesized by the well-known Schotten-Baumann reaction - the acylation of the corresponding amines with 2,2-dimethylpropionyl chloride. On the basis of FT-IR data for diluted solutions (concentrations lower than 10-3 mol dm-3) of the N-monosubstituted 2,2-dimethylpropanamides in carbon tetrachloride and dichloromethane, the exact position of the N-H stretching bands was established. The conformations and structures of N-monosubstituted 2,2-dimethy/propanamides were exactly proven on the basis of spectroscopic data. These results are in good accordance with 'H NMR and MS data.
AB  - U našem proučavanju amidne veze istraživali smo konformacije nekih N-monosupstituisanih 2,2-dimetilpropanamida opšte

formule CH3C(CH3)2CONHR, u kojima se VR mijenja od n-alkil (C1-C7), izo-alkil (C3-C6), sec-butil, alil, cikloalkil (C3-C6), fenil, do

4-supstituisanog fenila, (goje su supstituenti fenoro, metil, cijano, bromo, nitro, karboksimetil, N,N-dimetilamino, metoksi, trilluorometil)

2-feniletil acetofenil. Amidi su sintetizovani dobro poznatom Schotten-Baumanovom reakcijom - acilovanjem odgovarajućeg amina sa

2,2-dimetil propionil hloridom. Na bazi FT-IR podataka za razblazene rastvore (koncentracije nize od N-monosupstituisanih 2,2-dimetilpropanamida u CCI4 i CH2Cl2, ustanovijen je tačan položaj N-H stretching vipca. Konformacije i

strukture N-monosupstituisanih 2,2-dimetilpropanamida su egzaktno dokazane na bazi spektroskopskih podataka. Rezultati se dobro

slažu sa 'H NMR I MS podacima.

The amides were synthesized by the well-known Schotten-Baumann reaction - the acylation of the corresponding amines with 2,2-dimethylpropionyl chloride. On the basis of FT-IR data for diluted solutions (concentrations lower than 10-3 mol dm-3) of the N-monosubstituted 2,2-dimethylpropanamides in carbon tetrachloride and dichloromethane, the exact position of the N-H stretching bands was established. The conformations and structures of N-monosubstituted 2,2-dimethy/propanamides were exactly proven on the basis of spectroscopic data.
