Antioxidant and cytotoxic activity of fatty oil isolated by supercritical fluid extraction from microwave pretreated seeds of wild growing Punica granatum L.

AB  - Pomegranate seeds are an excellent source of compounds with biological activity. In the present study, the pomegranate seed oil (PSO) was tested for antioxidant and cytotoxic potential. PSO was extracted by super-critical fluid extraction with previous treatment of seeds by microwaves and analysed for its fatty acid profile, carotenoid and tocopherol contents. PSO demonstrated to be a good source of polyunsaturated fatty acids (PUFAs) (83.5%), followed by monounsaturated fatty acid (MUFA) and saturated fatty acid (SFA) (9.5% and 7.0%, respectively). Concerning fatty acid profile, punicic (C18:3), oleic (C18:1) and linoleic (C18:2) acids were the predominant ones. Carotenoids and tocopherols were identified and quantified by HPLC. Results showed that the PSO was a good source of gamma-tocopherol as dominant among investigated compounds (128.6 mg/100 g). The extract was screened for its antioxidant activity using alpha-TEAC assay. Cytotoxic effect was determined by MTT test against malignant HeLa, LS174 and A549 cells (IC50 = 49.51-91.54 mu g/mL), and against normal MRC-5 and EA.hy926 cells (IC50  gt  200 mu g/mL). The best effect was exhibited on cervix adenocarcinoma cell line while no cytotoxicity was found for normal cells pointing out PSO safety.

AB  - Silver/poly(N-vinyl-2-pyrrolidone) (Ag/PVP) nanocomposites containing Ag nanoparticles at different concentrations were synthesized using gamma-irradiation. Cytotoxicity of the obtained nanocomposites was determined by MU assay in monolayer cultures of normal human immunocompetent peripheral blood mononuclear cells (PBMC) that were either non-stimulated or stimulated to proliferate by mitogen phytohemagglutinin (PHA), as well as in human cervix adenocarcinoma cell (HeLa) cultures. Silver release kinetics and mechanical properties of nanocomposites were investigated under bioreactor conditions in the simulated body fluid (SBF) at 37 degrees C. The release of silver was monitored under static conditions, and in two types of bioreactors: perfusion bioreactors and a bioreactor with dynamic compression coupled with SBF perfusion simulating in vivo conditions in articular cartilage. Ag/PVP nanocomposites exhibited slight cytotoxic effects against PBMC at the estimated concentration of 0.4 mu mol dm(-3), with negligible variations observed amongst different cell cultures investigated. Studies of the silver release kinetics indicated internal diffusion as the rate limiting step, determined by statistically comparable results obtained at all investigated conditions. However, silver release rate was slightly higher in the bioreactor with dynamic compression coupled with SBF perfusion as compared to the other two systems indicating the influence of dynamic compression. Modelling of silver release kinetics revealed potentials for optimization of Ag/PVP nanocomposites for particular applications as wound dressings or soft tissue implants.

AB  - Silver doped hydroxyapatite (HAP) [Ca9.95Ag0.05(PO4)(6)(OH)(2)] composite coatings with natural polymer organosolv lignin (Lig) were produced by electrophoretic deposition (EPD) on titanium. Coatings were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), field emission scanning electron microscopy (FE-SEM), attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR) and X-ray photoelectron spectroscopy (XPS). The corrosion stability of electrodeposited coatings was evaluated in vitro in Kokubo's simulated body fluid (SBF) at 37 degrees C using electrochemical impedance spectroscopy (EIS). Antimicrobial properties are directly proportional to the rate of silver ions release from the coatings, determined from inductively coupled plasma spectrometry (ICP-AES). The obtained results are in good agreement with viability of pathogenic bacteria strain Staphylococcus aureus TL in suspension, which had completely disappeared after 24 h. Composite Ag/HAP/Lig coatings were confirmed as non-toxic for healthy immunocompetent peripheral blood mononuclear cells (PBMC).

AB  - Composite coatings based on lignin, obtained by electrophoretic deposition (EPD) on titanium, were investigated. The properties of HAP/Lig coatings were characterized using X-ray diffraction (XRD) analysis, scanning electron microscopy (SEM), thermogravimetric analysis (TGA), attenuated total reflection Fourier transform infrared spectroscopy (ATR-FT-IR) and X-ray photoelectron spectroscopy (XPS), as well as the MTT test of cytotoxicity. The results showed that higher lignin concentrations protected the HAP lattice during sintering, thereby improving the stability of the HAP/Lig coatings. The cell survival of peripheral blood mononuclear cells (PBMC) after proliferation indicates that the HAP/Lig coating with 1 wt% Lig electrodeposited on titanium was non-toxic with significant promise as a potential bone-repair material.
