AB  - Most chemical and physical methods employed in visualizing latent marks have shown detrimental effects on human health and, therefore, some research groups have directed their attention to the utilization of various (bio)polymers with the aforementioned purpose. Although chitosan is widely used in medicine, pharmacy, food, and drug delivery systems, there are very few studies that address this biopolymer utilization in forensic applications, such as the detection of latent fingerprints. We used chitosan crosslinked with sodium tripolyphosphate and conjugated with L-lysine to enhance the visualization of latent fingerprints, due to its ability to interact with fingerprint sweat residues. These conjugates were prepared using different (w/w) amounts of chitosan and tripolyphosphate (6/1; 4/1; 1/1; 1/4; and 1/6), and those with the most promising results (i.e., 6/1 formulation) were investigated in detail. Fourier transform infrared (FT-IR) spectroscopy confirmed interactions between components of the systems. Optical microscopy and scanning electron microscope (SEM) analysis showed that prepared powder formulations were uniform in size and confirmed that chitosan/tripolyphosphate/lysine conjugates bind easily to the sweat and lipid residues present in the latent fingerprints. The testing of prepared conjugates demonstrated the potential of these systems as bio-based powder substitution for commercially available powders.

AB  - Hidrogelovi na bazi poli(metakrilne kiseline) - PMAA imaju veliku primenu u sistemimaza ciljano otpuštanje aktivnih supstanci zato što su pH osetljivi, netoksični i biokompatibilni.Ipak zbog izrazite hidrofilnosti ovi hidrogelovi se mogu koristiti samo za kontrolisano otpuštanjehidrofilnih supstanci. U ovoj disertaciji predstavljena je modifikacija PMAA hidrogelovaamfifilnim supstancama: proteinom - kazeinom i fosfolipidnim nanočesticama - lipozomima,kako bi se dobio nosač slabo vodorastvorne aktivne supstance (SVAS). Opisana je sintezanosača, njihova karakterizacija primenom infracrvene spektroskopije sa Furijeovomtransformacijom (FTIR) i skenirajućom elektronskom mikroskopijom (SEM) i karakterizacijalipozoma primenom dinamičkog rasipanja svetlosti (DLS) i metodom elektroforetskog rasipanjasvetlosti (ELS). FTIR spektri su pokazali da su hidrofobne interakcije uspostavljene izmeđuSVAS model leka – kofeina i kazeina omogućile inkapsulaciju kofeina i njegovo kontrolisanooslobađanje, dok su prisutni lipozomi omogućili bolju kontrolu brzine oslobađanja kofeina.Ispitan je i uticaj parametara sinteze (stepen neutralizacije metakrilne kiseline, koncentracijaumreživača i kofeina, prisustvo itakonske kiseline i lipozoma, udeo i oblik lipozomneformulacije) na morfološke karakteristike nosača, njihovo bubrenje i kinetiku oslobađanjakofeina u dve sredine različitih pH vrednosti koje su simulirale uslove prisutne ugastrointestinalnom traktu čoveka. Praćenje oslobađanja kofeina iz nosača u ovim sredinamaizvršeno je primenom ultraljubičaste spektroskopije (UV). Izvedena je detaljna analiza dobijeniheksperimentalnih podataka otpuštanja kofeina u in vitro uslovima, primenom nekolikomatematičkih modela. Najbolja kinetika otpuštanja kofeina postignuta je iz nosača sa potpunoneutralizovanom metakrilnom kiselinom, 1,6mol% umreživača i inkorporiranim lipozomima ukojima je inkapsuliran kofein. Ovaj nosač je odabran za dalje analize - ispitano je njegovoponašanje u uslovima koji su simulirali put nosača kroz gastrointestinalni trakt čoveka. Rezultatisu pokazali da kofein može da se inkapsulira u izabrani nosač u koncentracijama koje su više odnjegove maksimalne rastvorljivosti i da se postigne njihovo ciljano otpuštanje. Inkapsulacijakofeina u koncentracijama ovih vrednosti, postignuta je dodatkom nikotin-amida koji sakofeinom gradi stabilni kompleks. Dobijeni rezultati pokazuju da sintetisani nosači imaju velikipotencijal za ciljanu dostavu slabo vodorastvotnih supstanci.
AB  - Hydrogels based on poly(methacrylic acid) (PMAA) have been extensively used insystems for targeted delivery of active substances because they are pH-sensitive, non-toxic andbiocompatible. However, being highly hydrophilic, these hydrogels are able to deliver onlyhydrophilic active substances. In this dissertation, modification of PMAA hydrogels withamphiphilic substances: protein - casein and phospholipidic nanoparticles - liposomes in order todesign a hydrophilic carrier for a poorly water-soluble substance (PWSS) are presented. Thesynthesis of the carriers and their characterization using the Fourier Transform InfraredSpectroscopy (FTIR) and the Scanning Electron Microscopy (SEM) and the characterization ofthe liposomes using the Dynamic Light Scattering (DLS) and the method of the ElectrophoreticLight Scattering (ELS) are described. The FTIR spectra showed that hydrophobic interactionswere established between PWSS model drug - caffeine and casein and enabled caffeineencapsulation in the carrier and its controlled release, while the presence of the liposomesenabled better control of caffeine release rate. The morphological characteristics of the carriers,their swelling behavior and the caffeine release kinetics were investigated depending on thechanges in the synthesis parameters (neutralization degree of methacrylic acid, crosslinker andcaffeine concentrations, introduction of itaconic acid and liposomes, volume ratio and form ofliposomal formulation) in two media with different pH values that simulated the environment inhuman gastrointestinal tract. Ultraviolet (UV) Spectroscopy was used to monitor caffeine releasefrom these carriers. Data from caffeine in vitro release experiments were analyzed in detail usingseveral mathematical models. The best kinetic of caffeine release was achieved form the carrierin which methacrylic acid was complitely neutralized and which had 1,6mol% of crosslinker andincorporated liposomes with encapsulated caffeine. The behavior of this carrier was furtherinvestigated in the environment which simulated the conditions in the human gastrointestinaltract. Results showed that this carrier could be used for the encapsulation of caffeine inconcentrations which were higher than its solubility and for targeted delivery of theseconcentrations. Encapsulation of caffeine in these values of concentration was achieved byadding nicotinamide, which forms stable complex with caffeine. Presented results showed thatsynthesized carriers have great potential for targeted delivery of poorly water-soluble substances.

Effect of Encapsulated Probiotic Starter Culture on Rheological and Structural Properties of Natural Hydrogel Carriers Affected by Fermentation and Gastrointestinal Conditions

AB  - The suitability of natural hydrogel carriers with probiotic starter culture as whey beverages supplements was examined by assessing their rheological and structural changes during the fermentation and gastrointestinal conditions. Effect of encapsulated cells on the carrier structure is of great importance for the selection of proper material for the preparation of functional supplements. The structural changes of the chitosan-coated alginate/whey carriers were considered based on (1) cell viability and the carrier average volume vs. time (2) the storage and loss modulus vs. time obtained under low oscillator strain conditions, (3) FTIR analysis and (4) SEM cross-sectional observation of the hydrogel carriers. The presence of chitosan coating and fermentation conditions increased cell viability up to 9.01 +/- 0.18 (log CFU/g). According to our results, the encapsulated cells induce weakening of carriers under the gastric conditions but improve their mechanical stability under the intestinal condition. The mechanical behaviour of carriers was also considered in order to formulate the rheological constitutive model equation for describing the irreversible structural changes under the gastric and intestinal conditions. The cell leakage under the gastric condition after the 2 h was less than 5%. Carriers are rapidly degraded under the intestinal condition which ensures the release of cells and provides their beneficial effects on the host health. Our results indicate that this type of coated carrier is suitable to be used for encapsulation of probiotic starter culture in the production of fermented whey-based products.

AB  - pH sensitive, nontoxic, and biocompatible poly(methacrylic) acid (PMAA) based soft networks have been extensively used in the design of systems for targeted drug delivery. Still, their highly hydrophilic nature limits their potential to be used as a carrier of poorly water-soluble substances. With the aim to overcome this limitation, the present study details a new approach for modification of PMAA based carriers using two amphiphilic components: casein and liposomes. The FTIR analysis revealed structural features of each component as well as the synergetic effect that originated from the formation of specific interactions. Namely, hydrophobic interactions between the poorly water-soluble model drug (caffeine) and casein enabled caffeine encapsulation and controlled release, while addition of liposomes ensured better control of the release rate. The morphological properties of the carriers, swelling behavior, and release kinetics of caffeine were investigated depending on the variable synthesis parameters (neutralization degree of methacrylic acid, concentration of caffeine, presence/absence of liposomes) in two different media simulating the pH environment of human intestines and stomach. The data obtained from in vitro caffeine release were correlated and analyzed in detail using several mathematical models, indicating significant potential of investigated carriers for targeted delivery and controlled release of poorly water-soluble substances.

AB  - Horseradish peroxidase (HRP) is a highly specific enzyme with great potential for use in the decolorization of synthetic dyes. A comprehensive study of HRP immobilization using various techniques such as adsorption and covalent immobilization on the novel carrier Purolite (R) A109 with a special focus on enzymatic decolorization and toxicity of artificially colored wastewater. The immobilized preparations with an activity of 156.21 +/- 1.41 U g(-1) and 85.71 +/- 1.62 U g(-1) after the HRP adsorption and covalent immobilization, respectively, were obtained. Stability and reusability of the immobilized preparations were also evaluated. A noteworthy decolorization level (similar to 90%) with immobilized HRP was achieved. Phytotoxicity testing using Mung bean seeds and acute toxicity assay with Artemia salina has confirmed the applicability of the obtained immobilized preparation in industrial wastewater plants for the treatment of colored wastewater.

AB  - This work is focused on immobilization of laccase from Myceliophthora thermophila expressed in Aspergillus oiyzae (Novozym 51003 (R) laccase) on amino modified fumed nano-silica (AFNS) and the possible use in bioremediation. Hereby, for the first time, factors affecting the immobilization of Novozym 51003 (R) laccase on AFNS were investigated for defining the immobilization mechanism and optimizing the utilization of AFNS as support for laccase immobilization. The highest specific activity (13.1 IU.mg(-1) proteins) was achieved at offered 160 mg per g of AFNS and for the same offered protein concentration the highest activity immobilization yield, reaching 68.3% after the equilibrium time, at optimum pH 5.0, was obtained. Laccase immobilization occurs by adsorption as monolayer enzyme binding in 40 min, following pseudo-first-order kinetics. The possible use of obtained immobilized preparation was investigated in degradation of pesticide lindane. Within 24 h, lindane concentration was reduced to 56.8% of initial concentration and after seven repeated reuses it retained 70% of the original activity.

AB  - Lipase-catalyzed transesterification of vitamin C by natural triglycerides is promising approach for cost-efficient synthesis of liposoluble food and cosmetic antioxidants. Nevertheless, application of these alternative acyl donors is insufficiently explored, despite of their low price, wide abundance and possibility for obtaining versatile products. Within current study, fatty acid ascorbyl esters were synthesized from vitamin C and natural triglycerides (lard, sunflower, coconut and linseed oil) in a process catalyzed by immobilized lipase Novozym (R) 435 and their controlled release was examined. High conversions were achieved at optimized conditions, even at high substrate concentrations, thus high concentrations of acsorbyl-ester were synthesized using these natural lipids. All ester mixtures exhibited very high capacity for scavenging of DPPH radicals, among which linseed oil derived esters, with ascorbyl linolenate as prevailing compound, were the most efficient (IC50 0.663 mu M), while coconut oil derived ester mixture (IC50 0.739 mu M), composed predominantly of molecules with medium side chains, was second best. Coconut oil derived esters were successfully incorporated in typical cosmetic formulations for controlled release of bioactive compounds - O/W emulsion and gel-emulsion. Franz cell diffusion study demonstrated that esters release from mixture and two carrier systems was formulation dependent and revealed decrease of effective diffusivities with ester side acyl chain length increase. According to calculated effective diffusivity coefficients, faster trans-membrane delivery of the same molecule was achieved from gel-emulsion comparing to O/W emulsion. Applied approach enabled highly efficient cost-saving production of fatty acid ascorbyl esters, which could find direct application in various lipophilic products.

AB  - Glycoderivatives are an important class of molecules with enormous relevance in numerous biological phenomena; therefore, they have a key role in the learning, understanding, and assessment of different diseases. Nanotechnology, and in particular the design of new nanomaterials, is one of the areas of greatest interest today. In this case, graphene nanomaterials represent very interesting platforms for studying glycosystems, glyconanomaterials that combine the biomolecular recognition and the characteristics of nanoscale objects in the development of early diagnosis systems, and efficient specific therapeutic modalities. In this mini-review, we discuss some results recently described in the literature on the conjugation of graphene materials and carbohydrates through the selective interaction of glycoenzymes in graphene to create new materials with biosensing applications, the development and application of sugar-graphene composites, and finally biosystems combining the properties of graphene with metallic nanoparticles and sugars for the creation of excellent glyconanomaterials as novel systems for the therapy or diagnosis of important diseases such as cancer or diabetes.

AB  - Immobilization of Lactobacillus rhamnosus ATCC7469 in poly(vinyl alcohol)/calcium alginate (PVA/Ca-alginate) matrix using "freezing-thawing" technique for application in lactic acid (LA) fermentation was studied in this paper. PVA/Ca-alginate beads were made from sterile and non-sterile PVA and sodium alginate solutions. According to mechanical properties, the PVA/Ca-alginate beads expressed a strong elastic character. Obtained PVA/Ca-alginate beads were further applied in batch and repeated batch LA fermentations. Regarding cell viability, L. rhamnosus cells survived well rather sharp immobilization procedure and significant cell proliferation was observed in further fermentation studies achieving high cell viability (up to 10.7 log CFU g(-1)) in sterile beads. In batch LA fermentation, the immobilized biocatalyst was superior to free cell fermentation system (by 37.1%), while the highest LA yield and volumetric productivity of 97.6% and 0.8 g L-1 h(-1), respectively, were attained in repeated batch fermentation. During seven consecutive batch fermentations, the biocatalyst showed high mechanical and operational stability reaching an overall productivity of 0.78 g L-1 h(-1). This study suggested that the "freezing-thawing" technique can be successfully used for immobilization of L. rhamnosus in PVA/Ca-alginate matrix without loss of either viability or LA fermentation capability.

A novel chitosan/tripolyphosphate/L-lysine conjugates for latent fingerprints detection and enhancement

AB  - Most chemical and physical methods employed in visualizing latent marks have shown detrimental effects on human health and, therefore, some research groups have directed their attention to the utilization of various (bio)polymers with the aforementioned purpose. Although chitosan is widely used in medicine, pharmacy, food, and drug delivery systems, there are very few studies that address this biopolymer utilization in forensic applications, such as the detection of latent fingerprints. We used chitosan crosslinked with sodium tripolyphosphate and conjugated with L-lysine to enhance the visualization of latent fingerprints, due to its ability to interact with fingerprint sweat residues. These conjugates were prepared using different (w/w) amounts of chitosan and tripolyphosphate (6/1; 4/1; 1/1; 1/4; and 1/6), and those with the most promising results (i.e., 6/1 formulation) were investigated in detail. Fourier transform infrared (FT-IR) spectroscopy confirmed interactions between components of the systems. Optical microscopy and scanning electron microscope (SEM) analysis showed that prepared powder formulations were uniform in size and confirmed that chitosan/tripolyphosphate/lysine conjugates bind easily to the sweat and lipid residues present in the latent fingerprints. The testing of prepared conjugates demonstrated the potential of these systems as bio-based powder substitution for commercially available powders.

AB  - The aim of this study was to develop functional edible films containing cocoa (Theobroma cacao L.) powder extract using alginate, pectin and chitosan in combination with proteins (whey protein isolate, soya and hemp protein). The films were examined for their physico-chemical (dry matter content, colour, thickness), mechanical (elongation at break (EAB)), bioactive (the content of total polyphenols (TPC), flavan-3-ols (F3olC), antioxidant capacity (AC)) and sensory properties. The plain alginate film exhibited the highest EAB (29.1%). The highest TPC and F3olC were determined in plain alginate (29 mg GAE/g and 2.75 mg (+)-catechin/g) and pectin (29 mg GAE/g and 2.25 mg (+)-catechin/g) films. The addition of proteins resulted in prolonged release of polyphenols and enhanced functional properties; however, the formation of protein-polyphenol complexes caused slight alterations in the bioactive composition of the films. The obtained results indicate a high potential of the developed films as functional, biodegradable form of active food packaging.

AB  - The electrospray process is a process that uses an electric field to influence the liquid dripping out of a capillary tube in order to obtain a reduction in the mean droplet diameter. The theory regarding this process is called electrohydrodynamics. Based on the previous experimental results and on the published theories, important variables are listed. Some of those variables have a significant influence on the droplet diameter, while others should be controlled in order to check the applicability of the mentioned theories. Since theoretical equations have a simple power-type form, that form was used for obtaining a correlation of particle diameters when alginate solutions are used. Previous experimental data and a Generalized Reduced Gradient (GRG) nonlinear solver were used for calculating the coefficients present in the correlation. Interesting results were obtained because certain variables were eliminated during the calculation. Based on the presented analysis, future experiments should monitor more variables and computational fluid dynamics (CFD) should also be used.

advantageous characteristics that make alginates so widely used are their
low cost, biocompatibility, low toxicity, and especially, easy gel forming in
mild conditions. In addition, an abundance of carboxyl and hydroxyl groups
enables easy chemical or biochemical modification which may give the
further improvement of their properties to tailor some specific demands, or
to eliminate some of their disadvantages. Biomedical applications of alginates mainly include encapsulations for drug delivery and controlled release,
tablet formulation, wound healing, as a scaffold for tissue engineering,
dental impression materials, water absorbents, chelators, and so on. Their
use in this field is attributed to their nonimmunogenicity and hydrophilicity, the simple formulation in any form (such as capsule, film, bead, or fiber),
and easy blending with other polymers. However, application of alginates
for modern biomedical treatments demand a comprehensive understanding
of their properties, broad knowledge about the relationship between their
structure and their functions, as well as, clear insight in possibilities for their
chemical or biochemical methods of modifications. This chapter is focused
on recent advances related to the applications, research, and development
of the biomedical use of alginate and survey of possible chemical and
biochemical means for their modifications.

AB  - Three dimensional polymer networks, hydrogels, have shown a great potential in
controlled release and targeted delivery of active substances. pH sensitive hydrogels
based on poly(methacrylic acid) have attracted significant attention due to their
biocompatibility, non-toxicity and the possession of the huge number of ionogenic
carboxylic groups. On the other hand, the usage of these hydrogels for controlled release
and targeted delivery of poorly water-soluble drugs is limited by their highly hydrophilic
nature. We overcame this limitation by modifying PMAA with amphiphilic substance -
casein. The addition of this natural protein enabled the encapsulation, targeted delivery
and controlled release of poorly water-soluble model drug - caffeine. Three carriers with
different neutralization degree of methacrylic acid were synthesized via free radical
polymerization. The FTIR spectra revealed that two types of interactions were
established between casein and caffeine: hydrophobic interactions and hydrogen bonds.
The SEM micrographs showed that the structure of the carriers depended on the change
of neutralization degree of the methacrylic acid. The swelling of synthesized carriers and
the caffeine in vitro release were monitored in two environments at 37°C: 0.1M HCl
pH=1.2 (simulation of environment in human stomach) and phosphate buffer pH=6.8
(simulation of environment in human intestines). The synthesized carriers had higher
swelling degree and the higher release rate of caffeine in the phosphate buffer pH=6.8
than in 0.1M HCl. The increase in neutralization degree of methacrylic acid caused the
increase in swelling degree of the carriers and the increase in release rate of caffeine.
Presented results showed that synthesized carriers are promising candidates for targeted
delivery of poorly water-soluble drugs.

AB  - Chaga (Inonotus obliquus) is a parasitic fungus, which has been used in traditional medicine in Russia and other northern European countries in the treatment of cancer, gastrointestinal andliver diseases. It has been a subject of intensive researchrecently, confirming many of its health-beneficial effects. In order to obtain a product that would allow modified and prolonged release of the Chaga's active metabolites, hot water Chaga extract was immobilized using calcium - alginate. The extract, which was predominantly composed of carbohydrates (57 %), also contained a relatively high amount of antioxidants/phenolic compounds (130 mg gallic acid equivalents per g of dry extract) and exhibited pronounced radical scavenging activity. It showed significant antibacterial activity as well, inhibiting growth of tested bacterial strains at concentrations of 1.25-20 mg/mL. Entrapment efficiency was about 80 %, and the extract-alginate system showed pH-dependant extract release; there was negligible release at pH 1.75 (gastric pH), and the release gradually increased with the increase in pH, reaching similar to 43 % of immobilized extract at pH 8.5 after 90 min. Such a product could be used as a dietary supplement, adjuvant in therapy of gastrointestinal diseases or as a food additive.

AB  - Puffballs are fungi that produce globose fruiting bodies that undergo autolysis, transforming their insides into a spore bearing, powdery mass. Mature fruiting bodies are traditionally used to treat open skin wounds. In this study, methanol extracts of two puffball species, Handkea excipuliformis and Vascellum pratense, were examined and compared in order to provide insight into the changes these mushrooms undergo during maturation, with respect to their potential use in skin care and wound treatment. Some compounds involved in skin care and regeneration were quantified, and it was found that maturation increases the concentrations of almost all of these compounds. Antioxidant activity was also more pronounced in mature fruiting body extracts, which was in correlation with the higher content of antioxidants. Tyrosinase inhibition was vastly improved with autolysis, correlating with the higher phenolic content in mature fruiting body extracts. Antimicrobial activity was negatively affected by autolysis in the case of H. excipuliformis, whereas autolysis had little effect on the antimicrobial activity of V. pratense. Autolysis generally improved the biological activity and increased the concentrations of compounds involved in skin care, which justifies the traditional use of puffballs and makes them good candidates for various potential cosmetic and medicinal skin-care products.

AB  - Methanol extracts of mosaic puffball (Handkea utriformis, Bovistella utriformis, Lycoperdon utriforme, Calvatia utriformis - current name is a subject of debate), from three different stages - mycelium (HUMIC), immature (HUI) and mature fruiting bodies (HUM) were characterized and tested for antioxidant, antimicrobial and inhibitory activity on tyrosinase, acetyholinesterase (AChE), butyrylcholinesterase (BuChE) and 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA-R). Immature, edible, fruiting bodies were shown to be a good source of antioxidants (11.5 mg/g of extract) and cholesterol-lowering agent, lovastatin (234 mu g/g of extract), and exhibited significant antimicrobial activity. In addition, HUI showed good and selective AChE (4.48 mg/mL) and non-lovastatin related HMG-CoA-R inhibition (1.16 mg/mL), which all together suggests that regular consumption of it may have health benefits. Mature fruiting bodies, inedible due to powdery consistence, have been used in traditional medicine for wound treatment; their extract was relatively rich in free ergosterol (31.65 mg/g of extract), N-acetylglucosamine (24 mg/g of extract), alpha-tocopherol (4 mg/g of extract) and had best overall antioxidant activity, which was in correlation with its highest phenolic content (19.4 mg GAE/mL). It also exhibited significant tyrosinase inhibitory activity (0.22 mg/mL) and thus could be used in medicinal and cosmetic products for wound healing and bleaching. Mycelium, which can be easily grown in laboratory conditions did not have the same properties as, neither immature or mature fruiting bodies, although it showed prominent antimicrobial activity, notably against Pseudomonas aeruginosa (MIC = 0.0625 mg/mL) and could be a source of antimicrobial compounds.

AB  - The aim of the present study was the optimization of the extraction conditions (particle size, solid-to-solvent ratio, extraction time and procedure) via total polyphenol content and radical scavenging activity, using wild thyme (Thymus serpyllum L.) as a natural source of polyphenols. According to the total polyphenols yield, particle size, solvent-to-solid ratio, and extraction technique have shown statistically significant influence, whereas the exposure time has not been a relevant factor. In all three extraction procedures (maceration, heat-and ultrasound-assisted extraction), the highest polyphenols yield was reached using the smallest plant particles and the highest solvent-to-solid ratio. Moreover, polyphenols content was significantly higher after ultrasound-assisted extraction, in comparison to the extraction at room and high temperature. According to the antioxidant recovery, the influence of different extraction parameters varied depending on the applied antioxidant tests and extraction procedures. 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)-ABTS radical scavenging capacity of extracts followed the next trend: maceration and ultrasound-assisted extraction  gt  heat-assisted extraction , while in 2,2-diphenyl-1-picrylhydrazyl-DPPH neutralization: ultrasound-assisted extraction  gt  heat-assisted extraction  gt  gt  maceration. The study represents an important step in the formulation of antioxidant-rich extracts with potential use in food, pharmaceutical, and cosmetic industry.

AB  - In the present study, spray-dried Thymus serpyllum extracts obtained by using four extraction
techniques (maceration, heat-assisted extraction, extraction in ultrasound bath and extraction by
ultrasound probe) were characterized through determination of total polyphenols and total
flavonoids contents, antioxidant capacity, zeta potential, bulk density and solubility. Polyphenols
yield was statistically higher in the extract obtained by ultrasound probe, in comparison to the
extracts prepared using maceration, heat-assisted extraction and extraction in ultrasound bath.
On the other hand, extraction technique did not show statistically significant influence on the
flavonoids concentration. Furthermore, extraction procedures did not show significant effect on
antioxidant neutralization of DPPH radicals, while flavonoids yield had correlation with the
DPPH radical-scavenging capacity assay. However, the extracts obtained using maceration and
ultrasound waves (ultrasound bath) have shown significantly better antioxidant capacity in the
preservation of β-carotene, compared to the extracts prepared by using high temperature and
ultrasound probe. The extract obtained in maceration has shown statistically better stability
(higher absolute value of zeta potential), compared to the extracts prepared using high
temperature and ultrasound waves. According to the results of Pharmacopoeia test, bulk density
of spray-dried T. serpyllum extracts was ranked by significance in the following order: heatassisted extraction>ultrasound bath and ultrasound probe>maceration. Relatively high
solubility of spray-dried extracts has been found in the following order: maceration>ultrasound
bath and ultrasound probe>heat-assisted extraction. Spray-dried T. serpyllum extract prepared
using maceration represents a powder with satisfying physicochemical properties, as well as a
good source of substances with significant antioxidant activity, particularly in emulsions, which
enable its use in functional food and pharmaceutical products.

AB  - The main object of study is application of whey protein concentrate (WPC) and whey protein hydrolysate (WPH) for probiotic encapsulation. The controlled enzymatic hydrolysis was applied to change technological properties of WPC what leads to obtained different carriers. Probiotic carriers (beads) were made by electrostatic extrusion. Bead properties (mechanical properties, FTIR fingerprint, cell release) and parameters of fermentation were examined. According to cell release, it can be concluded that WPH build less porous matrix with alginate than WPC. Beads with WPH contained more living cells and suffered more changes during fermentation than beads with WPC. Probiotic viability in simulated gastrointestinal conditions (SGIC) is the most critical parameter for probiotic encapsulation. Probiotic encapsulated in protein-alginate beads survived SGIC with more than 96% viable cells, compared to 37.43% for free culture. According to all examined parameters, it can be concluded that WPH builds more suitable carrier for probiotic culture than WPC.

AB  - Probiotic cultures are becoming more popular in human nutrition primarily because modern consumers tend to use food products that have a more positive impact on their health. The therapeutic effect of food with added live microorganisms (probiotics), especially milk products, is promoted by health professionals. In order to achieve the desired therapeutic effect of food product, probiotic bacteria have to enter the body in appropriate number and maintain viability during production and storage conditions. After consuming the product, it is important that an adequate number of cells survive conditions in the gastrointestinal tract. Preparation of a natural hydrogel carrier, which will enable the protection of the probiotic starter culture during production, storage and the gastrointestinal conditions was goal of this doctoral dissertation. Aim was full potential impact of functional food product on human health. Thesis analyzes two types of results: characterization of obtained carriers with probiotic starter culture using different encapsulation techniques and application of the carriers in preparation of functional food product. AB  - Probiotske kulture su sve više zastupljene u ljudskoj ishrani pre svega zbog težnje savremenih potrošača da konzumiraju kvalitetne prehrambene proizvodе koji imaju i pozitivan uticaj na ljudsko zdravlje. Terapeutski efekat hrane sa dodatim živim mikroorganizmima (probioticima) posebno na primerima mlečnih proizvoda se sve više promoviše od strane zdravstvenih radnika. Probiotske bakterije, da bi ostvarile željeni terapeutski efekat, neophodno je da se u organizam unesu u odgovarajućem broju i da se održi njihova vijabilnost tokom proizvodnog procesa i skladištenja proizvoda. Isto tako, posle konzumiranja proizvoda, potrebno je da adekvatan broj ćelija preživi uslove u gastro-intestinalnom traktu. β‐sitosterol and gentisic acid loaded1,2‐dipalmitoyl‐sn‐glycero‐3‐phosphocholine liposomal particles

AB  - The aim of the present study was the examination of the impact of β-sitosterol and gentisic acid on the characteristics of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine(DPPC) liposomal particles: (a) bilayer permeability (fluorescence spectroscopy),(b) particle size, polydispersity index (PDI) and zeta potential (photon correlation spectroscopy) and (c) thermal properties (differential scanning calorimetry).β-sitosterol induced the increase of liposomal bilayer rigidity, due to rearranging of the phospholipid chains, while gentisic acid enhanced the membrane fluidity, due to the reduced orderliness and the increase of phospholipid dynamics. The inclusion of β-sitosterol in liposomes caused a significant increase in particle diameter and PDI, while the encapsulation of gentisic acid did not have influence on particle size distribution. Apart from that, the presence ofβ-sitosterol resulted in the significant zeta potential increase, and thus a better stability of liposomal spheres(in the absence and in the presence of gentisic acid). β-sitosterol decreased main transition temperature (Tm) and phase transition enthalpy (∆H), and caused the disappearance of the pre-transition peak as well, whereas the presence of gentisic acid produced a slight decrease in Tm and increase of ∆H. Therefore, gentisic acid had more favourable, stabilizing interactions with phospholipids thanβ-sitosterol. Thus, it can be concluded that β-sitosterol is located in the bilayer interior between phospholipids acyl chains, and gentisic acid is incorporated near the outer leaflet of the phospholipid membrane, next to the polar head groups.β-sitosterol and gentisic acid loaded DPPC liposomal particles have a potential to be used in food and pharmaceutical products, due to the important individual and possible synergistic beneficial health properties ofβ-sitosterol and gentisic acid.
