The impact of pH value and temperature on polyphenol recovery, antioxidant capacity and physical properties of Serpylli herba's waste extracts

AB  - In the present study, Serpylli herba's waste extracts were prepared using 50% ethanol (pH 6) and 50%
ethanol with hydrochloric acid (pH 1.2), using maceration (25 °C) and heat-assisted extraction (HAE,
80 °C). The best extraction conditions were determined via analyzing total polyphenol (TPC) and
flavonoid contents (TFC), antioxidant capacity, extraction yield, zeta potential, density, and surface
tension. In maceration and HAE, TPC were 1.18±0.06 and 1.43±0.05 mg GAE/mL (pH 6), and
0.69±0.05 and 0.86±0.02 mg GAE/mL (pH 1.2). The same trend is noticed in the case of TFC:
0.33±0.01 and 0.42±0.02 mg CE/mL (pH 6) and 0.24±0.01 and 0.27±0.01 mg CE/mL (pH 1.2). ABTS
radical scavenging activity was 775.0±3.5 and 623.8±5.1 mmol Trolox/mL (pH 6), and 269.2±3.1
and 327.9±7.7 mmol Trolox/mL (pH 1.2). In DPPH assay, IC50 was 1.64±0.04 and 1.48±0.03 mg/mL
(pH 6), and 7.59±0.07 and 5.11±0.09 mg/mL (pH 1.2). Extraction yield was 0.541±0.049 and
0.583±0.019% (pH 6), and 0.938±0.057 and 0.889±0.060% (pH 1.2). Zeta potential was -2.58±0.02
and -2.08±0.10 mV (pH 6), and 1.49±0.37 and 1.08±0.09 mV (pH 1.2). Density was 0.945±0.001
and 0.949±0.001 g/mL (pH 6), and 1.008±0.001 and 1.010±0.001 g/mL (pH 1.2). Surface tension
was 29.2±0.3 and 30.5±1.0 mN/m (pH 6), and 32.6±2.3 and 36.5±2.3 mN/m (pH 1.2). The best
properties were determined in the extract obtained in HAE at pH 6. The aim of this study is to provide
evidence of the physico-chemical properties of Serpylli herba's waste extracts that can add value and
improve the quality of existing food, pharmaceutical, and cosmetic products.

AB  - A new micro-kinetic model of the enzyme-catalyzed synthesis of fructo-oligosaccharides (FOS) was developed. A commercial enzyme mixture Pectinex® Ultra SP-L derived from Aspergillus aculeatus was used. A variety of initial enzyme concentrations (1–5 vol%) and sucrose concentrations (400–600 g/L) were experimentally investigated and included in kinetic modeling. Several variations of kinetic mechanisms and corresponding models have been examined. A hybrid genetic algorithm was used to predict the kinetic parameters simultaneously for all experimental data. The best fitting model has been adopted, and with an average error of 13.34%, it describes the experimental data very well. The influence of initial concentrations on the conversion of sucrose and production of FOS is being carefully investigated. It was shown that the initial sucrose concentration significantly affects the highest level of FOS concentration, but the enzyme concentration controls the time at which maximum is reached as well as the rate of FOS decomposition.

AB  - A new micro-kinetic model is proposed for galacto-oligosaccharide (GOS) synthesis with β-galactosidase from Aspergillus oryzae. Several kinetic models with similar reaction mechanisms, i.e., steps and kinetic constants, have been investigated covering a range of initial lactose (20-40 wt %) and enzyme concentrations (0.5-2 mg/mL). The kinetic parameters were estimated simultaneously for all experimental results using a hybrid genetic algorithm. A detailed analysis of the effect of process parameters on the concentrations of lactose and GOS is performed. The proposed model has a very good quantitative fit of the experimental data, with an average error of 9.34%. More importantly, qualitative trends for the change in the concentration of lactose and GOS are predicted excellently, especially if one takes into account the variation of both concentrations of enzyme and lactose in a range wider than reported in the literature to date. Also, the analysis between the two comparative reactions of transglycosylation and lactose hydrolysis confirmed the positive effect of increasing lactose concentration on the production of GOS. On the other hand, a higher enzyme concentration resulted in a faster production of GOS but with more intensive decomposition at longer reaction times.

AB  - Food technology has adopted new principles and practices that are rapidly changing the food sector. New foods are now available under more uniform standards and better quality control. Globalised food market offers opportunities for manufacturers to increase production and profit, and at the same time, consumers benefit from the choice of food products like never before. All this is possible only because of the innovations in the food sector. One of such innovations is encapsulation technology, which aims to preserve food quality, enhance the sensorial properties of food and increase the efficiency in food processing.

This book discusses the uses of encapsulation technology in food practices and conventional processes and also highlights new directions in food processing. In the introductory chapters’ review of encapsulation technologies, carrier materials and criteria for their selection, analytical methods for characterisation of encapsulated products and some aspects of product design and process optimisation. The most important achievements of encapsulation technology in the food sector are reviewed in the later chapters related to encapsulation of food ingredients, food biocatalysts and examples of usage of encapsulated active ingredients in the dairy and meat industry, beverage production, etc. In addition, the implementation of nanotechnology in the food sector is reviewed, emphasizing the most important materials and technologies for the production of nanoencapsulates. The book is a valuable source of information on encapsulation technology, for academia and industry, especially the food sector, with the aim of enhancing knowledge transfer.

Pharmacological potential of Thymus serpyllum L. (wild thyme) extracts and essential oil: A review

AB  - In traditional medicine Thymus serpyllum L. (wild thyme) herb represents a part of a large number of herbal medicinal formulations such as syrups, tinctures, infusions, teas, and decoctions. In recent years, there has been a growing interest in testing the biological properties of wild thyme, because the plant is a high-quality raw material, rich in essential oil and pharmacologically active polyphenolic compounds, which can be included in various formulations in the pharmaceutical, cosmetic, food, and chemical industries. Wild thyme extracts and essential oil have shown significant nitric oxide, ABTS and DPPH radicals scavenging potential, ferrous ion chelating property, reducing activity and ability to inhibit peroxidation of deoxyribonucleic acid (DNA), proteins and lipids. Wild thyme extracts exerted antibacterial activity against Bacillus cereus, Enterococcus faecalis, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella enterica, Yersinia enterocolitica, as well as Lactic acid bacteria. Wild thyme essential oil has shown antibacterial potential against E. coli, L. monocytogenes, P. aeruginosa, S. enteritidis, S. aureus, Streptococcus salivariu, S. mutans, S. sanguinis, S. pyogenes, E. feacalis, B. cereus, B. subtilis, B. pumilis, Lactobacillus acidophilus, Proteus mirabilis, Klebsiella pneumoniae, Salmonella choleraesuis, and Salmonella poona. T. serpyllum extract possesses antifungal activity against Fusarium solani, F. moniliforme, Aspergillus flavus, Microsporum canis, Alternaria species, Candida albicans, and C. glabarata. The essential oil efficiently inhibited the growth of C. albicans, C. glabrata and Aspergillus fungi. Depending on the concentration, wild thyme extract significantly relaxed spontaneous contractions, as well as acetylcholine-, potassium chloride-, barium chlorideand calcium chloride-induced contractions of the isolated rat ileum. The extract significantly reduced the expression of the inducible enzyme cyclooxygenase-2 and inhibited acetylcholinesterase, myeloperoxidase and α-glucosidase activity. T. serpyllum extract has shown cytotoxic activity on human breast cancer cell lines, while essential oil has shown the antitumor potential in human cell lung cancer, colon, cervical, hepatocellular, prostate, and breast adeno-carcinoma.

In recent years, there has been a growing interest in testing the biological properties of wild thyme, because the plant is a high-quality raw material, rich in essential oil and pharmacologically active polyphenolic compounds, which can be included in various formulations in the pharmaceutical, cosmetic, food, and chemical industries. Wild thyme extracts and essential oil have shown significant nitric oxide, ABTS and DPPH radicals scavenging potential, ferrous ion chelating property, reducing activity and ability to inhibit peroxidation of deoxyribonucleic acid (DNA), proteins and lipids. Wild thyme extracts exerted antibacterial activity against Bacillus cereus, Enterococcus faecalis, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella enterica, Yersinia enterocolitica, as well as Lactic acid bacteria. Wild thyme essential oil has shown antibacterial potential against E. coli, L. monocytogenes, P. aeruginosa, S. enteritidis, S. aureus, Streptococcus salivariu, S. mutans, S. sanguinis, S. pyogenes, E. feacalis, B. cereus, B. subtilis, B. pumilis, Lactobacillus acidophilus, Proteus mirabilis, Klebsiella pneumoniae, Salmonella choleraesuis, and Salmonella poona. T. serpyllum extract possesses antifungal activity against Fusarium solani, F. moniliforme, Aspergillus flavus, Microsporum canis, Alternaria species, Candida albicans, and C. glabarata. The essential oil efficiently inhibited the growth of C. albicans, C. glabrata and Aspergillus fungi. Depending on the concentration, wild thyme extract significantly relaxed spontaneous contractions, as well as acetylcholine-, potassium chloride-, barium chlorideand calcium chloride-induced contractions of the isolated rat ileum. The extract significantly reduced the expression of the inducible enzyme cyclooxygenase-2 and inhibited acetylcholinesterase, myeloperoxidase and α-glucosidase activity. T. serpyllum extract has shown cytotoxic activity on human breast cancer cell lines, while essential oil has shown the antitumor potential in human cell lung cancer, colon, cervical, hepatocellular, prostate, and breast adeno-carcinoma.",

AB  - Thymus serpyllum L (thyme) aqueous extract was encapsulated within three different types of particles. Thus, electrostatic extrusion was applied to obtain alginate, alginate/chitosan, and alginate/inulin particles with encapsulated extract which is known as a rich source of polyphenolic compounds. The effect of admixed chitosan and inulin was evaluated from the point of polyphenols release by using Franz diffusion cell. The alginate/chitosan particles exhibited the effective diffusion coefficient of 1.94 center dot 10(-9) center dot m(2) s(-1) which was about 2.7 times lower in comparison to that of alginate/inulin particles and about 6.3 times lower than the effective diffusion coefficient of plain alginate particles. Swelling studies also confirmed decreased permeability of composite particles compared to plain alginate particles. In addition, according to the Korsmeyer-Peppas model, the composite particles exhibited anomalous diffusion of polyphenols controlled by molecular diffusion and polymer relaxation. Practical applications The calcium alginate particles are meant for delivery of biologically active compounds in foods with added value. Coating of alginate particles using chitosan has been investigated to control release of encapsulated biologically active compounds, but also as an improver of the particles structure.

Lipid nanocarriers for phytochemical delivery in foods

AB  - The delivery of different bioactive compounds in foods is a popular and convenient approach to provide healthy functional foods, but it usually requires an additional step—encapsulation—to provide functional foods with maintained efficacy of active compounds. For that purpose, the usage of matrix originated from natural sources, which are biocompatible and biodegradable, is the demand of customers; therefore, lipid nanocarriers are favored compared to polymeric carrier,s and they have been extensively studied over the last decades. This chapter aims to offer the current state-of-the art in lipid nanocarriers technology for phytochemicals delivery in foods. The main focus is given to solid lipid nanoparticles, nanostructured lipid carriers, lipid nanoemulsions, and nanoliposomes. Better control of drug release in comparison to more frequently used delivery systems, delayed rate of degradation, possibility to evade biotoxicity of carriers, and possibility of industrial scale production assert the solid lipid nanoparticles as a promising tool to successfully deliver phytochemicals. Nanostructured lipid carriers are considered as the “new generation” of solid lipid nanoparticles. Due to the different domains in the structure of nanostructured lipid carriers, they enable better retention of actives and higher encapsulation efficiency and reduce the possibility of burst effect during storage, thus providing extensive physical stability. Lipid nanoemulsions are also interesting due to the fact that they can be added into foodstuff without compromising the sensorial properties of the food. Finally, nanoliposomes are being increasingly used in food and beverage industries to deliver various antioxidant and antimicrobial agents from plant sources. Anyway, more extensive research has to be conducted to define the digestion and metabolism pattern of nanocarriers not only to ensure optimal consumption levels and appropriate product formulations, but also to analyze the costs of the industrial scale production.

(PDI) i zeta potencijal lipozoma, pripremljenih metodom tankog filma i
upotrebom fosfatidilholina soje, Phospholipon 90 (Ph). Merenjem
polarizacije (p) i anizotropije (r), u temperaturnom intervalu od 20 do 60˚C,
upotrebom fluorofore 1,6-difenil-1,3,5-heksatrien (DPH), pokazano je da
povećanjem koncentracije holesterola u lipozomima dolazi do smanjenja
fluidnosti i povećanja rigidnosti membrane lipozoma. Foton korelaciona
spektroskopija je kod malih unilamelarnih lipozoma (SUV) pokazala
kontinuirano povećanje veličine čestica sa porastom koncentracije
holesterola, dok kod multilamelarnih lipozoma (MLV) trend kontinuiranog
rasta čestica nije primećen. U oba slučaja, statistički značajno veće čestice
su dobijene sa najvišom koncentracijom holesterola (50 mol %). Prema
dobijenim vrednostima PDI, samo SUV bez holesterola i sa 5, 30 i 40 mol %
holesterola, poseduju usku raspodelu čestica po veličini (PDI manji od 0,2).
Vrednosti zeta potencijala, kao važnog parametra stabilnosti sistema, bile su
negativne u svim uzorcima, dok su najviše apsolutne vrednosti dostignute sa
50 mol % holesterola. Optimalna koncentracija holesterola za dobijanje
lipozoma odgovarajuće stabilnosti i fluidnosti membrana jeste 50 mol %.
AB  - The aim of this study was examination of the influence of various cholesterol
content (0-50 mol %) on membrane fluidity, vesicle size, polydispersity index
(PDI) and zeta potential of liposomes prepared using the thin film method
and phosphatidylcholine from soybean, Phospholipon 90 (Ph). Polarization
(p) and anisotropy (r) measurements, within the temperature range of 20 to
60˚C, using the fluorescent probe 1,6-diphenyl-1,3,5-hexatriene (DPH), have
shown that increasing the cholesterol content in the liposomes decreased the
fluidity and enhanced the rigidity of liposomal membranes. The photon
correlation spectroscopy data have shown that the size of small unilamellar
vesicles (SUV) increased continuously with the increase in cholesterol
concentration, while multilamellar vesicles (MLV) did not show similar
trend. In both cases, significantly larger vesicles were obtained using the
highest content of cholesterol (50 mol %). According to the PDI values, only
SUV without cholesterol and with 5, 30 and 40 mol % of cholesterol exhibit
narrow size distribution (PDI < 0,2). Furthermore, values of zeta potential,
as an important parameter of system stability, were negative in all samples,
while the highest absolute values were achieved with 50 mol % of cholesterol. The optimal concentration of cholesterol to obtain stabile
liposomes and an appropriate fluidity of the membrane has been 50 mol %.

AB  - Liposomes are known as convenient carriers for a broad range of actives. The release rate of actives can be adjusted/controlled by the addition of different surfactants. The aim of this study was to compare the effects of two surfactants, Span 20 and Span 40, on liposomes release properties. In particular, the study shows how the membrane modification affects the mass transfer resistances and liposome size compared with the common liposomes. The aqueous extract of thyme was used as a model substance. It is a valuable source of natural antioxidants - polyphenols. The diffusion of polyphenols from native thyme extract (un-encapsulated) and from different types of liposomes was conducted using Franz diffusion cell. The results were approximated with Fick's second law and they implied that liposomes modified with Span 20 provided the slowest release of polyphenolic compounds due to the highest value of mass transfer resistance (2.523 x 10(6) s/m).

The diffusion of polyphenols from native thyme extract (un-encapsulated) and from different types of liposomes was conducted using Franz diffusion cell. The results were approximated with Fick's second law and they implied that liposomes modified with Span 20 provided the slowest release of polyphenolic compounds due to the highest value of mass transfer resistance (2.523 x 10(6) s/m).",

AB  - The paper reports about the preparation and characterization of hydrogel particles containing liposomes loaded with resveratrol as an active compound. The materials used for preparation of the particles were chosen to be suitable for food industry. Different polymer concentrations affect particles shape, size, size distribution, as well as the release kinetics of resveratrol. The diameter of particles varied from 360 to 754 μm, while the narrow size distribution was observed for all types of particles. Release studies were performed in Franz diffusion cell and the results showed the prolonged release of resveratrol from all samples, but the sample with the highest content of polymer (2.5% w/w) in particular stood out. The research provides useful information about liposomes containing active compound encapsulated in hydrogel matrices and offers the basis for its application in the food industry.
AB  - Ovaj rad daje podatke o pripremi i karakterizaciji čestica koje sadrže lipozome sa inkapsuliranom aktivnom komponentom resveratrolom. Komponente koje ulaze u sastav ovih čestica odabrane su tako da mogu jednostavno da se primene u prehrambenoj industriji. Prikazan je uticaj različitih koncentracija početnih rastvora polimera čija upotreba je dozvoljena u hrani, a samim tim i njihove viskoznosti na veličinu formiranih čestica, njihov oblik i raspodelu veličina, ali i na otpuštanje resveratrola iz ovih složenih sistema. Prečnik čestica bio je između 360 i 754 μm, dok je uska raspodela veličina detektovana u svim uzorcima. Otpuštanje resveratrola praćeno je u Francovoj difuzionoj ćeliji gde su rezultati ukazali na produženo oslobađanje resveratrola u svim uzorcima. Ipak, uzorak koji je imao najveći udeo polimera u početnom rastvoru (2,5% w/w) najsporije je otpuštao aktivnu komponentu. Ovi rezultati daju korisne podatke o kompleksnim sistemima gde je aktivna komponenta inkapsulirana u lipozome dalje obložena polimerom čime doprinose potencijalnoj aplikaciji ovih i sličnih sistema u prehrambene proizvode.

Diffusion of polyphenols from microparticles obtained from different encapsulation techniques

AB  - Polyphenols are compound which are widely studied because of their antioxidative and potential therapeutic properties. Systems for the controlled release of drugs offer a number of benefits comparing with traditional forms of medicines and because of that these systems are widely researched. Encapsulation of Thymus serpyllum L. aqueous extract within Ca-alginate beads was studied in order to produce dosage formulations containing polyphenolic compounds. Electrostatic extrusion was applied for encapsulation of thyme aqueous extract in alginate gel beads. In addition to hydrogel beads, heat-dried and freeze-dried forms of beads were examined. The obtained beads with approximately 2 mg g-1 of gallic acid equivalents encapsulated in 0.015 g ml-1 of alginate were uniformly sized spheres of about 730 μm. Encapsulation efficiency varied in the range 80-98 % depending on the encapsulation method. Besides, the analysis reveals that the encapsulation process and the material used did not degrade the bioactive compounds, as the total antioxidant content remained unchanged. This was verified by the FTIR analysis proving the absence of chemical interactions between extract compounds and alginate. Addition of a filler substance such as sucrose and inulin in the dried product reduced its collapse and roundness distortion during drying process. Also, the objective of this paper is to investigate the possibility of using liposomes as carriers of polyphenols and influence the membrane composition on the release rate of encapsulated polyphenols. Experiments show how the membrane modification affects the mass transfer comparing to a conventional liposomes. Liposomes were modified with surfactants Span 20, Span 40, Tween 20 and Tween 60, and thyme tea extract was used as a source of polyphenols. The diffusion of polyphenols from thyme extract, dispersion of conventional liposomes and liposomes modified with Span 20, Span 40, Tween 20 and Tween 60 was studied using Franz diffusion cell. AB  - Sistemi za kontrolisano otpuštanje aktivnih supstanci predstavljaju istraživačku oblast koja se intenzivno proučava zbog niza prednosti koje ovi sistemi pružaju i mogućeg unapređenja dosadašnjih, tradicionalnih formi lekova. Vodeni ekstrakt biljke majčine dušice je inkapsuliran u alginatne čestice, kao i alginat-hitozan i alginat-inulin čestice elektrostatičkom ekstruzijom. Pored hidrogel čestica, ispitivane su i suve i liofilisane forme. Takođe, fosfolipidne mikročestice (lipozomi) su se pokazale potencijalno pogodnim nosačima polifenolnih jedinjenja u sistemima ovog tipa. Eksperimenti su izvedeni korišćenjem Franz–ove difuzione ćelije, u kojima je praćena difuzija polifenola iz: čajnog ekstrakta, alginatnih čestica, alginat-hitozan, alginat-inulin čestica, disperzije konvencionalnih lipozoma, disperzije lipozoma modifikovanih Span-om 20, Span-om 40,Tween-om 20 i disperzije lipozoma modifikovanih Tween-om 60. Obradom eksperimentalnih rezultata određeni su koeficijenti difuzije za svaki od sistema, kao i odgovarajući difuzioni otpori. Dobijene alginatne čestice su bile veličine oko 730 μm, dok su lipozomi bili u opsegu od 350 do 900 nm. Stepen inkapsulacije za alginante čestice je bio visok, preko 90 %. Osim toga, analiza je pokazala da postupak inkapsulacije nije degradirao polifenolna jedinjenja, jer je ukupan sadržaj antioksidanasa ostao nepromenjen. Odsustvo hemijskih interakcija između ekstrakta čaja i alginata je dokazano i FTIR analizom. Jedan od predmeta ovog rada je ispitivanje uticaja sastava membrane lipozoma na brzinu oslobađanja inkapsuliranih polifenola. Eksperimentalna ispitivanja treba da pokažu kako modifikacija membrane lipozoma utiče na prenos mase, u odosu na konvencionalne lipozome. Lipozomi su modifikovani površinski aktivnim materijama Span 20, Span 40, Tween 20 i Tween 60. Takođe, na osnovu dobijenih rezultata se može zaključiti da se inkapsulacijom polifenola u fosfolipidne mikročestice – lipozome, značajno usporava njihova difuzija, a da se modifikacijom membrane može postići dodatno usporavanje prenosa mase, što zavisi od strukture molekula kojim se modifikacija vrši. Molekuli Tween-a 20 su se pokazali uspešnijim modifikatorima membrane lipozoma od molekula Tween-a 60. Otpor koji membrana modifikovana Tween-om 20 pruža difuziji polifenola je veći i dovodi do 4.93 puta sporije difuzije u odnosu na sistem sa neinkapsuliranim polifenolima...

AB  - Wild Thyme (Thymus serpyllum) is perennial, herbaceous plant of Lamiaceae family. It has
aromatic, antioxidant, antiseptic, anthelmintic, repelent, antirheumatic, expectorant,
antispasmotic, carminative, sedative and diuretic properties. Optimization of the extraction by
maceration of T. serpyllum was carried out through varying degrees of fragmentation (0.3, 0.7
and 1.5) and the type of solvent (water, ethanol and mixtures of those in weight ratio 50:50 and
70:30). Extraction efficiency was expressed via total polyphenols content and antioxidant
activity. The highest total polyphenols content and antioxidant activity were recorded in 50%
ethanolic extract, with a degree of fragmentation of 0.3, 921,51 mg GA/g DW and 0,538 mM
Trolox/g DW, respectively. No significant differences were observed between total polyphenols
and antioxidant activity in water and in 70% ethanolic extracts. The lowest total polyphenols
content and antioxidant activity were obtained in 96% ethanolic extract (70.73 mg GA/g DW and
0,142 mM Trolox/g DW, respectively).

Systems for the controlled release of drugs offer a number of benefits comparing with traditional forms of medicines and because of that these systems are widely researched. The objective of this paper is to investigate the possibility of using liposomes as carriers of polyphenols and influence of the membrane composition on the release rate of encapsulated polyphenols. Experiments show how the membrane modification affects the mass transfer comparing to a conventional liposomes. Liposomes were modified with surfactants Tween 20 and Tween 60, and thyme tea extract was used as a source of polyphenols. The diffusion of polyphenols from thyme extract, dispersion of conventional liposomes and liposomes modified with Tween 20 and Tween 60 were studied using Franz diffusion cell. From the experimental data diffusion coefficients were determined for each of the systems, as well as the corresponding diffusion resistances. From the obtained results it can be concluded that the encapsulation of polyphenols in liposomes significantly slows diffusion, and with membrane modification can be achieved further slowing. The diffusion resistance of the liposome membrane modified with Tween 20 and Tween 60 is about 5 times higher compared to the diffusion of unencapsulated polyphenols from the thyme extract.
AB  - Sistemi za kontrolisano otpuštanje lekova predstavljaju istraživačku oblast koja se intenzivno proučava zbog niza prednosti koje ovi sistemi pružaju i mogućeg unapređenja dosadašnjih, tradicionalnih formi lekova. Fosfolipidne mikročestice (lipozomi) su se pokazale potencijalno pogodnim nosačima aktivnih supstanci u sistemima ovog tipa. Predmet ovog rada je ispitivanje uticaja sastava membrane lipozoma na brzinu oslobađanja inkapsuliranih polifenola. Eksperimentalna ispitivanja su pokazala kako modifikacija membrane lipozoma utiče na prenos mase, u odnosu na konvencionalne lipozome. Lipozomi su modifikovani površinski aktivnim materijama Tween 20 i Tween 60, dok je kao izvor polifenola korišćen čajni ekstrakt biljke majčine dušice. Eksperimenti su izvedeni korišćenjem Franz-ove difuzione ćelije, u kojima je praćena difuzija polifenola iz: čajnog ekstrakta, disperzije konvencionalnih lipozoma, disperzije lipozoma modifikovanih pomoću Tween 20 i disperzije lipozoma modifikovanih pomoću Tween 60. Obradom eksperimentalnih rezultata određeni su koeficijenti difuzije za svaki od sistema, kao i odgovarajući difuzioni otpori. Na osnovu dobijenih rezultata može se zaključiti da se inkapsulacijom polifenola u fosfolipidne mikročestice-lipozome, značajno usporava njihova difuzija, a da se modifikacijom membrane može postići dodatno usporavanje prenosa mase, što zavisi od strukture molekula kojim se modifikacija vrši. Molekuli Tween 20 kao i Tween 60 su se pokazali uspešnim modifikatorima membrane lipozoma. Otpori koje membrane modifikovane navedenim polisorbatima pružaju difuziji polifenola su veći i dovode do 4,9 puta sporije difuzije u odnosu na sistem sa neinkapsuliranim polifenolima.

Encapsulation of thyme (Thymus serpyllum L.) aqueous extract in calcium alginate beads

AB  - BACKGROUND: Encapsulation of Thymus serpyllum L. aqueous extract within calcium alginate beads was studied in order to produce dosage formulations containing polyphenolic compounds. Electrostatic extrusion was applied for encapsulation of thyme aqueous extract in alginate gel beads. In addition to hydrogel beads, heat-dried and freeze-dried forms of beads were examined. METHODS: Encapsulation systems were examined and compared in order to choose the optimal one with respect to entrapment efficiency, preservation of antioxidant activity and thermal behaviour under heating conditions simulating the usual food processing. RESULTS: The beads obtained with approximately 2 mg g(-1) of gallic acid equivalents encapsulated in 0.015 g mL(-1) of alginate were spheres of a uniform size of about 730 mu m. Encapsulation efficiency varied in the range 50-80% depending on the encapsulation method. Besides, the analysis reveals that the encapsulation process and the material used did not degrade the bioactive compounds, as the total antioxidant content remained unchanged. This was verified by Fourier transform infrared analysis, which proved the absence of chemical interactions between extracted compounds and alginate. Addition of a filler substance, such as sucrose and inulin, in the dried product reduced its collapse and roundness distortion during drying process. CONCLUSION: This study demonstrates the potential of using hydrogel material for encapsulation of plant poplyphenols to improve their functionality and stability in food products.

Isolation of Hemoglobin from Bovine Erythrocytes by Controlled Hemolysis in the Membrane Bioreactor

AB  - In this work, we describe an optimized procedure based on gradual hemolysis for the isolation of hemoglobin derived from bovine slaughterhouse erythrocytes in a membrane bioreactor. The membrane bioreactor system that provided high yields of hemoglobin (mainly oxyhemoglobin derivate) and its separation from the empty erythrocyte membranes (ghosts) was designed at a pilot scale. Ten different concentrations of hypotonic media were assessed from the aspect of the extent of hemolysis, hematocrit values of the erythrocyte suspensions, cell swelling, and membrane deformations induced by decreased salt concentration. Effective gradual osmotic hemolysis with an extent of hemolysis of 88% was performed using 35 mM Na-phosphate/NaCl buffer of pH 7.2-7.4. Under these conditions most of the cell membranes presented the appearance of the normal ghosts under phase contrast microscope. The hemoglobin purity of  gt 80% was confirmed by SDS-PAGE. Kinetic studies showed that maximal concentration of hemoglobin was reached after 40 min, but the process cycle at which recovery of 83% was achieved lasted for 90 min. The dynamics of both steps, (1) transport through the membrane of erythrocytes during process of hemolysis and (2) transport through the reactor filters, were evaluated.

AB  - Dugi niz godina, i kod nas i u svetu, razvijaju se tehnološki postupci za izolovanje strukturno očuvanog i biološki aktivnog hemoglobina koji bi mogao da se koristi kao zamenik za krv, kao izvor biološki visokoaktivnog hemskog gvožđa u prevenciji anemije kod ljudi i životinja, ili kao reagens u dijagnostici. U ovom radu optimizovan je postupak za izolovanje hemoglobina iz eritrocita poreklom iz otpadne klanične goveđe krvi sa ciljem dobijanja preparata hemskog gvožđa za prevenciju anemije kod životinja. Testirana je osetljivost goveđih eritrocita na osmotsku lizu sa ciljem definisanja optimalnog puferskog sistema za efikasno izvođenje kontrolisane hemolize. Dobijeni rezultati su pokazali da goveđi eritrociti imaju povećanu osmotsku osetljivost u odnosu na humane eritrocite, a da je optimalan puferski sistem za izvođenje njihove kontrolisane hemolize 35 mM natrijum-fosfatni/NaCl pufer pH 7,2-7,4. Kontrolisana hemoliza sa optimizovanim puferskim sistemom je izvedena u membranskom reaktorskom sistemu i ostvaren je prinos hemoglobina od 83±12%. Tokom ovog procesa nije došlo do oštećenja membrane eritrocita, a intaktne membrane praznih eritrocita bi se mogle koristiti za inkapsulaciju biološki aktivnih supstanci.

AB  - In this study the encapsulation of raspberry leaf, hawthorn, ground ivy, yarrow, nettle and olive leaf extracts was performed by electrostatic extrusion in alginate-chitosan microbeads, with ascorbic acid being used for the dissolution of chitosan. The original and encapsulated plant extracts were characterized for their polyphenol content and composition, mineral content and antioxidant capacity. Raspberry leaf encapsulating microbeads exhibited the highest total phenol content and antioxidant capacity, followed by hawthorn, while olive leaf microbeads contained the lowest total phenol content. High encapsulation efficiency was obtained for all extract encapsulating microbeads (80-89%). Nettle extract-containing microparticles were characterized with the largest particle size and irregular shape, due to a high content of microelements (copper, strontium, and zinc), which affected the geling process of alginate. Although the antioxidant stability of hydrogel microcapsules was deteriorated during refrigerated storage, which might be attributed to the instability of ascorbic acid, the obtained microbeads deliver significant biological activity and antioxidant potential which may increase the daily intake of antioxidants when implemented in a food product.

Diffusion of lidocaine hydrochloride from lipid microparticles

AB  - Lipid particles, as drug carriers, are of increasing research interest, because a sustained drug release and avoidance of side effects could be achieved by using them. Lidocaine hydrochloride is a very efficient local anesthetic, but it has a short effect. The objective was to prolong the effect of the drug by encapsulating lidocaine hydrochloride in phospholipids microparticles. Two different procedures were used for the preparation of phospholipids microparticles. In both cases, phosphatidylcholine and lysophosphatidylcholine were used for the preparation of microparticles in which 5 % w/w lidocaine hydrochloride solution was encapsulated. The standard Franz diffusion cell was used for the experimental diffusion rate determination. The obtained results show that diffusion rate from microparticles is significantly lower than from the lidocaine hydrochloride solution, which means that this kind of microparticles could be used for the prolonged drug release. There was no much difference in diffusion rates from microparticles obtained by different procedures. That indicates that only the composition of the particles membrane has an influence on the lidocaine hydrochloride release rate.
