Dual responsive hybrid hydrogels for controlled release of local anesthetic

AB  - Intelligent hydrogels, such as pH sensitive hydrogels based on 
poly(methacrylic acid) (PMAA) are widely used for targeted drug 
delivery. Still, PMAA lack of good mechanical properties often 
limits its application. In order to overcome this limitation nano cellulose (NC) was extracted from wood waste material and then 
added to PMAA because NC is biocompatible, non-toxic and has 
excellent mechanical properties. Further, carboxymethyl cellulose 
(CMC) (cellulose derivate widely used for controlled release of 
drugs) was added. CMC can stabilize magnetite nanoparticles (MN) 
which is then also added. MN can significantly improve mechanical 
properties of hydrogels and also possess magnetic properties due to 
which MN can be used for targeted drug delivery. The as-prepared 
material can protect drug, deliver it to the site of action, control its 
release rate and enable in that way its efficient application with re duced side effects. Local anesthetic - lidocaine hydrochloride (LH) 
is often administrated by injection which can induce severe side ef fects. This problem is solved in present study by encapsulating LH 
into hydrogels based on PMAA, NC, CMC and MN (PMNC/MN L). PMNC/MN-L hydrogels were characterized by FTIR and SEM 
spectroscopies and single compressive tests and then their swelling 
behavior and LH release were analyzed. Present study offers unique 
approach for green synthesis of dual responsive hydrogels with im proves properties and their application for controlled release of local 
anesthetic with reduced side effects.

AB  - This work was focused on the synthesis of polyurethane networks (PUNs) based on polycaprolactone (PCL) as soft segment (SS) and Boltorn (R) aliphatic hyperbranched polyester of the second pseudo generation and 4,4 '-diphenylmethane diisocyanate (MDI) as the parts of the hard segments (HS), by a two-step solution polymerization. In order to find the best ratio between HS and SS to achieve good thermal and mechanical properties, suitable hydrophobicity and morphology of the PUNs, various experimental analyses were conducted. The obtained results revealed that features of the PUNs highly depend on the PCL content. The increase in the PCL content leads to the increase in thermal stability, hydrophobicity and appearance of the microphase separation, but on the other side, it also leads to the decrease in storage modulus in the rubbery plateau, crosslinking density and glass transition temperatures of PUNs. This work shows that features of PUNs can be easily adjusted for a specific application by careful selection of the SS and HS ratio.

AB  - pH sensitive, nontoxic, and biocompatible poly(methacrylic) acid (PMAA) based soft networks have been extensively used in the design of systems for targeted drug delivery. Still, their highly hydrophilic nature limits their potential to be used as a carrier of poorly water-soluble substances. With the aim to overcome this limitation, the present study details a new approach for modification of PMAA based carriers using two amphiphilic components: casein and liposomes. The FTIR analysis revealed structural features of each component as well as the synergetic effect that originated from the formation of specific interactions. Namely, hydrophobic interactions between the poorly water-soluble model drug (caffeine) and casein enabled caffeine encapsulation and controlled release, while addition of liposomes ensured better control of the release rate. The morphological properties of the carriers, swelling behavior, and release kinetics of caffeine were investigated depending on the variable synthesis parameters (neutralization degree of methacrylic acid, concentration of caffeine, presence/absence of liposomes) in two different media simulating the pH environment of human intestines and stomach. The data obtained from in vitro caffeine release were correlated and analyzed in detail using several mathematical models, indicating significant potential of investigated carriers for targeted delivery and controlled release of poorly water-soluble substances.

AB  - A series of environmentally friendly polyurethane (PU) networks based on polycaprolactone as soft segment (SS) and Boltorn (R) aliphatic hyperbranched polyester of the second pseudo generation as crosslinking agent was prepared by a two-step polymerization in solution. The chemical structure of the PU networks was analyzed by Fourier transform infrared spectroscopy, while XRD analysis revealed that these samples are amorphous. Swelling measurements, water absorption study, dynamic mechanical analysis, differential scanning calorimetry, thermogravimetric analysis, mechanical tests and scanning electron microscope revealed that, swelling behavior, hydrophobicity, thermal and mechanical features and morphology of PU networks show clear dependence on polycaprolactone content. The increase of the SS content induced appearance of the microphase separated morphology, increase of thermal stability, elongation at break, hydrophobicity and swelling ability in tetrahydrofuran and toluene, but at the same time it is responsible for the decrease of crosslinking density, glass transition temperatures, rigidity, tensile strength and Young's modulus of PU networks. Scanning electron microscope images of the fracture surfaces of PU networks show that their surface is rough, irregular, with flake-like structure, where size of flakes increases with increasing SS content. These results indicate that by choosing adequate SS content, PU coatings with diverse features based on polycaprolactone and Boltorn (R) aliphatic hyperbranched polyester can be designed for appropriate and desired application.

AB  - Soft polymeric networks based on poly(methacrylic
acid) (PMAA) are attractive candidates for targeted and
controlled drug release due to their non-toxicity,
biocompatibility and pH-sensitivity. The highly hydrophilic
nature of PMAA networks enables transport of hydrophilic
drugs only. This limitation has been overcome in present work
by a PMAA modification with casein and liposomes. Casein is a
natural amphiphilic protein which enabled the encapsulation
and targeted and controlled release of the model drug- caffeine.
The FTIR spectra showed that the hydrophobic interactions and
hydrogen bonds were established between the casein and
caffeine. The caffeine in vitro release was monitored in two media
at 37C: phosphate buffer pH=6.8, which simulated the pH
environment in the human intestines and 0.1M HCl pH=1.2,
which simulated the pH environment in the human stomach. The
presence of liposomes with the encapsulated caffeine in the
carriers caused the decrease in the speed of caffeine release.
Introduction of itaconic acid (IA) as a hydrophilic and pHsensitive substance with two carboxylic groups resulted in a nonregular structure of the carriers with large voids which caused
an increase in swelling rate of the carriers and an increase in
speed of caffeine release. All obtained results showed that the
targeted and controlled release of a poorly water-soluble
substance was achieved.

acid) (PMAA) are attractive candidates for targeted and
controlled drug release due to their non-toxicity,
biocompatibility and pH-sensitivity. The highly hydrophilic
nature of PMAA networks enables transport of hydrophilic
drugs only. This limitation has been overcome in present work
by a PMAA modification with casein and liposomes. Casein is a
natural amphiphilic protein which enabled the encapsulation
and targeted and controlled release of the model drug- caffeine.
The FTIR spectra showed that the hydrophobic interactions and
hydrogen bonds were established between the casein and
caffeine. The caffeine in vitro release was monitored in two media
at 37C: phosphate buffer pH=6.8, which simulated the pH
environment in the human intestines and 0.1M HCl pH=1.2,
which simulated the pH environment in the human stomach. The
presence of liposomes with the encapsulated caffeine in the
carriers caused the decrease in the speed of caffeine release.
Introduction of itaconic acid (IA) as a hydrophilic and pHsensitive substance with two carboxylic groups resulted in a nonregular structure of the carriers with large voids which caused
an increase in swelling rate of the carriers and an increase in
speed of caffeine release. All obtained results showed that the
targeted and controlled release of a poorly water-soluble
substance was achieved.",

AB  - The design of complex polymeric materials with predefined task-specific properties meets challenging issues: to establish synthesis-structure-properties relationship, as well as to understand the arrangement and interactions of the involved components. Hybrid networks of protein (casein) and polyelectrolyte (poly(methacrylic acid)) were synthesized and their swelling, dynamic-mechanical and morphological properties were investigated as functions of the neutralization degree of methacrylic acid and the concentrations of caseinate and crosslinker. The domination of different interactions between components led to diverse forms of casein macromolecules-from micelles to unfold chains. That resulted in very different structures and thereby properties of derived hybrid networks: from porous unswellable to highly swellable and reinforced ones. A great opportunity to easily modulate the final characteristics of the reported hybrid system to fit the need of specific application just through the change of one synthesis parameter was thereby demonstrated.

AB  - The aim of this study was the development of novel polysaccharide based films intended to be used as edible food packaging material. The films were prepared by solution casting method using highly methoxylated pectin (PEC) and poly(ethylene glycol) (PEG) of various molecular weights (400, 600 and 1000 gmol(-1)) in different ratios (5:1, 3:1 and 1:1). The film formation was supported by hydrogen bonding between PEC and PEG, which was evidenced by means of ATR-FTIR and NMR analysis. TGA revealed that generally PEG behaves like a pro-degrading agent for pectin, except in the case of PEC/PEG film with a ratio of 1:1. Furthermore, DSC thermograms indicated that PEG1000 exists as a separate phase in the pectin matrix while the formulations with PEG400 and PEG600 showed mainly amorphous morphology. The addition of PEG enhanced the plasticization of PEC films, as evidenced by progressive decreasing of the glass transition temperature values (T-g). The tensile test measurements showed that increasing concentration of PEG produced weaker and more flexibile films. Due to the increased molecular mobility, the pectin phase became more permeable to water vapor as the PEG concentration increased. The obtained results showed that the combination of both polymers resulted in interesting bio -inspired edible films with the potential to compete with commercially used synthetic package materials.

AB  - The sustainable tomorrow for future generations lies with the present industrial development toward the proper utilization of various bio-based products. For a transition to a higher level of sustainability, it is necessary to form a new platform for advanced technology products. This paper reports the development of new fully bio-based unsaturated polyesters resins (UPRs). A series of prepolymers were synthesized by varying saturated diacids (oxalic, succinic and adipic acid), itaconic acid and 1,2-propandiol. Dimethyl itaconate was used as a reactive diluent (RD) in amounts of 30, 35 and 40 wt%. Rheological measurements showed that the obtained resins possessed viscosities (234-2226 mPa s) amenable to a variety of liquid molding techniques. The impact of composition variables-prepolymer structure and amount of RD-on the chemical, mechanical and thermal properties of the thermosets was examined by DMA, TA and tensile measurements and was discussed in detail. The tensile properties (37-52 MPa), glass transition temperature (60-97 A degrees C) and coefficient of thermal expansion (71-168 10(-6) A degrees C-1) of the cured resins were in the desired range for UPRs. This investigation showed that UPRs based on itaconic acid can be tailored during synthesis of the prepolymer to meet the needs of different property profiles.

AB  - Present study reports synthesis and physico-chemical evaluation of hydrophobically modified pectin derivatives, obtained by reacting of pectin with di-acyl chlorides (glutaryl and sebacoyl chloride). Depending on length of the inserted carbon chains, the acylation resulted in possible formation of mono-grafted (isolated chains) and bi-grafted (chemical gels) structures. The structural features of obtained derivatives were investigated using FTIR spectroscopy, confirming the successful synthesis. The concentrated aqueous solutions of modified pectin showed interesting rheological properties, having lower values of apparent viscosity compared to neat pectin. Since the GPC analysis indicated that no degradation occurred, the viscosity decrease was explained by more heterogeneous organization within modified pectin solutions (microparticles together with sticky polymer entanglement). A shift in particle size distribution proved that proposed modifications also affected pectin solution properties in diluted regime. AB  - Goals. The goal of this study was a development of biocompatible composite hydrogels, structurally similar to native bone tissue, by incorporation of ~60 wt % of calcium hydroxyapatite (HA) into a matrix of hydrogels. Also, a possibility to control swelling kinetic and equilibrium swelling degree (SDeq) of hydrogels, by altering the degree of neutralization of the precursor (DN) was examined. Methods. Composite hydrogels, based on HA and poly(methacrylic acid) (PMAA), were synthesized by free-radical polymerization with different DN. Theoretical content of HA in synthesized composites was 60 wt %. Composites were synthesized by methods of dynamic mechanical analysis and scanning electron microscopy. SDeq and swelling kinetic were examined in distilled water and simulated body fluid. Results. Morphological observations revealed uniform distribution and strong bond of spherical HA particles within the polymer matrix. Swelling analyses demonstrated that SDeq is directly proportional to DN, while rheological examinations indicated inverse proportion between DN and storage modulus, but due to the HA particles inclusion, mechanical properties of composites were significantly better compared to monophasic PMAA hydrogels. Significance. Simple method of synthesis of composite hydrogels with high content of filler nanoparticles is presented. Incorporation of HA nanoparticles significantly improved mechanical properties of hydrogels, while at the same time was demonstrated a possibility to control swelling kinetic by influencing the DN.

Takođe, ispitan je uticaj neutralizacije prekursora polimerne mreže na kinetiku bubrenja i ravnotežni stepen bubrenja (SDeq).Metode - Kompozitni hidrogelovi, sastavljeni od HA i poli(metakrilne kiseline) (PMAA), sintetisani su slobodno-radikalskom polimerizacijom, pri različitim stepenima neutralizacije prekursora (SN). Svi sintetisani kompoziti sadrže visok maseni udeo HA (~60 mas %). Kompoziti su okarakterisani metodama skenirajuće elektronske mikroskopije (SEM) i dinamičko-mehaničke analize (DMA). SDeq i kinetika bubrenja ispitivani su u destilovanoj vodi i simuliranoj telesnoj tečnosti (SBF). Rezultati - Morfološka ispitivanja pokazala su uniformnu raspodelu i dobru povezanost sferičnih čestica apatita sa polimernom matricom. Analiza procesa bubrenja pokazala je proporcionalnu zavisnost SN i SDeq kompozita. Reološka ispitivanja ukazala su na obrnuto proporcionalnu zavisnost između SN i vrednosti modula sačuvane energije, koji su iznosili 55130 Pa u slučaju potpune neutralizacije i 77480 Pa u slučaju 0% neutralizacije. Značaj - Predstavljena je jednostavna metoda za sintezu kompozitnih hidrogelova sa visokim masenim udelom nanočestica punioca. Inkorporacijom nanočestica HA postignuto je značajno poboljšanje mehaničkih svojstava hidrogelova, dok je istovremeno pokazano da je promenom SN moguće kontrolisati kinetiku bubrenja hidrogelova. .

AB  - For the preparation of fully biobased unsaturated polyester resins (UPRs), the replacement of styrene with alternate nonpetroleum-based monomers turned out to be one of the most challenging tasks. Its complexity lies in the fact that reactive diluents (RD) have to have low viscosity and volatility, good compatibility with prepolymer, and capability to homopolymerize and copolymerize with its unsaturations. In this context, we directed our efforts to develop fully biobased UPRs using the dialkyl itaconates as an alternative to styrene. Therefore, a series of 100% biobased UPRs were prepared from itaconic acid and 1,2-propandiol and diluted by dialkyl itaconates. The resins were characterized by Fourier transform infrared spectroscopy, NMR, volatility, and viscosity measurements, while the cured samples were characterized by dynamic mechanical properties, thermomechanical analysis, thermogravimetric analysis data, and tensile tests. The influence of RD structure on the properties of cured samples was discussed in detail. It was shown that the prepared resins had evaporation rates of dialkyl itaconates of several orders of magnitude less compared to styrene. The cured resins with dimethyl itaconate showed comparable or even better thermal and mechanical properties compared to the one with styrene. This investigation showed that itaconic acid and dialkyl itaconates are promising bioresources for the preparation of fully biobased UPRs for mass consumption.

Cross-linking of highly methoxylated pectin with copper: the specific anion influence

AB  - The aim of this work was to investigate the influence of specific anions on the cross-linking process of highly methoxylated pectin using the following copper salts CuSO4; Cu(C2H3O2)(2); CuCl2; and Cu(NO3)(2) wherein the initial salt concentrations were varied from 0.5 up to 10 g l(-1). It was found that the anions affected the sorption capacity wherein the Cu2+ sorption capacity from the sulphate solution was the highest, while it decreased in the presence of CH3COO-, Cl- and NO3- ions, respectively. This difference was mostly pronounced at the higher initial salt concentrations (co(Cu2+)  gt  2 g l(-1)). The obtained beads were characterized by FTIR, MS, SEM/EDS microanalysis and mechanical compression tests up to 80% of deformation. The sorption data were applied to the Langmuir and Freundlich isotherm models and various calculated parameters confirmed the sulphate anion supportive nature in metal ion binding. The value of the K-f parameter for the cross-linking process of pectin in the presence of acetate, chloride and nitrate was approximately the same (K-f approximate to 0.027 g g(-1)), while it was higher in the presence of sulphate anions by more than 20% (K-f = 0.0352 g g(-1)). The predicted 1/n values (1/n  lt  1, 1/n  gt  1, 1/n = 1 for the sulphate, nitrate and acetate, and chloride anions, respectively) were the quantitative confirmation of the specific interactions involved in the cross-linking mechanism caused by different anions. The established anion influence was in accordance with the typical ion-specific influence on macromolecules in aqueous systems proposed by Hofmeister.

AB  - This study investigates a wide range of clinically relevant mechanical properties of poly(methyl methacrylate) (PMMA) denture base materials modified with di-methyl itaconate (DMI) and di-n-butyl itaconate (DBI) in order to compare them to a commercial PMMA denture base material. The commercial denture base formulation was modified with DMI and DBI by replacing up to 10 wt% of methyl methacrylate (MMA) monomer. The specimens were prepared by standard bath curing process. The influence of the itaconate content on hardness, impact strength, tensile, and thermal and dynamic mechanical properties was investigated. It is found that the addition of di-n-alkyl itaconates gives homogenous blends that show decreased glass transition temperature, as well as decrease in storage modulus, ultimate tensile strength, and impact fracture resistance with increase in the itaconate content. The mean values of surface hardness show no significant change with the addition of itaconates. The magnitude of the measured values indicates that the poly(methyl methacrylate) (PMMA) denture base material modified with itaconates could be developed into a less toxic, more environmentally and patient friendly product than commercial pure PMMA denture base material.

AB  - The aim of this work was to examine the possibility of modification of commercial denture base materials with itaconic acid esters, in order to obtain materials with lower toxicity and higher biocompatibility. Despite their relatively higher price compared to methacrylates, itaconic acid and itaconates are materials of choice for environmentally friendly applications, because they are not produced from petrochemical sources, but from plant products. A commercial system based on poly(methyl methacrylate) was modified using ditetrahydrofurfuryl itaconate (DTHFI), whereby the ratio of DTHFI was varied from 2.5 to 10 % by weight. Copolymerization was confirmed using FTIR spectroscopy, while SEM analysis showed the absence of micro defects and pores in the structure. The effects of the itaconate content on the absorption of fluids, the residual monomer content, thermal, dynamic-mechanical and mechanical properties (hardness, toughness, stress and elongation at break) were investigated. It was found that the addition of DTHFI significantly reduced the amount of residual methyl methacrylate, which made these materials less toxic. It was shown that increasing the DTHFI content resulted in materials with decreased glass transition temperatures, as well as with decreased storage modulus, ultimate tensile strength and impact fracture resistance; however the mechanical properties were in the rang prescribed by ADA standards, and the materials could be used in practice. The deterioration in mechanical properties was therefore worthwhile in order to gain lower toxicity of the leached monomer.

Adsorption of azo dyes on polymer materials

AB  - The use of polymeric adsorbents for the removal of azo dyes from solution has been reviewed. Adsorption techniques are widely used to remove certain classes of pollutants from waters, especially those which are not easily biodegradable. The removal of azo dyes as pollutants from wastewaters of textile, paper, printing, leather, pharmaceutical and other industries has been addressed by the researchers. The wider use of already available adsorbents is restricted due to their high costs which lead to investigation and development of new materials that can be cheaper, eficient and easy regenerated. The aim of this article is to present to the readers the widespread investigations in recent years of synthetic and natural polymers as adsorbents and potential replacement of conventional adsorbents. This review presents only the data obtained using raw, hydrogel, grafted and crosslinked forms of synthetic and nature based polymers, and the discussion is limited to these polymer-based materials and their adsorption properties.
AB  - Intenzivan industrijski razvoj praćen je sve većom količinom otpadnih voda, što u smislu zaštite životne sredine i održivog razvoja nalaže potrebu poboljšanja postojećih i uvođenje novih postupaka obrade otpadnih voda. Posebnu naučno-tehnološku pažnju zahtevaju azo boje, koje su veoma toksične i u prirodi veoma teško razgradive. Azo boje su podložne bioakumulaciji, a zbog kancerogenih i mutagenih svojstava neretko su pretnja zdravlju ljudi i očuvanju okoline. Primenu fizičko-hemijskih metoda za uklanjanje azo-boja iz otpadnih voda često ograničavaju visoke cene, potrebe za odlaganjem nastalog štetnog mulja ili nastanak toksičnih sastojaka razgradnje. Adsorpcija je jedna od najzastupljenijih metoda za uklanjanje raznih zagađivača iz otpadnih voda zbog jednostavnosti, ekonomičnosti i efikasnosti ove metode. S obzirom da su konvencionalni adsorbenti, kao što je aktivni ugalj, veoma skupi, u poslednje vreme se sve više ulaže u ispitivanja i razvoj novih materijala za uklanjanje zagadjivača iz otpadnih voda koji će biti lako razgradivi, netoksični, jeftini i efikasni u njihovom uklanjanju. U ovom radu biće prikazan pregled iz literature polimernih materijala za uklanjanje azo boja iz otpadnih voda. Polimerni materijali pokazuju prednosti u odnosu na konvencio-nalne adsrobente zbog jednostavne upotrebe i rukovanja s njima i mogućnosti oblikovanja u pogodne i željene forme. Adsorpcija na polimernim materijalima je najčešće reverzibilan proces, što nije slučaj kod konvencionalnih adsorbenta. Adsorbovana boja na polimernim materijalima, u zavisnosti od tipa boje, se lako može desorbovati u kiseloj ili alkalnoj sredini, kao i u acetonu, tako da se materijal može regenerisati i ponovo koristiti za uklanjanje boja iz otpadnih voda. U ovom radu će biti prikazani sintetički i prirodni polimeri u formi hidrogela, umreženih i kalemljenih sistema za uklanjanje azo boja. Takodje, prikazan je efekat različitih parametara kao što su koncentracija, pH i temperatura rastvora boje, na adsorpciju na polimernim materijalima.
