AB  - Tissue engineering strives for innovative solutions in addressing challenges associated with contemporary bone tissue implants. This study focuses on the electrophoretic deposition (EPD) of hydroxyapatite-based bioceramic composites containing antibacterial agents onto titanium surfaces. Two composite coatings, hydroxyapa-tite/chitosan (HAP/CS) and hydroxyapatite/chitosan/gentamicin (HAP/CS/Gent) were developed to combat issues such as poor adhesion, limited antibacterial potential, limited bioactivity, and potential toxicity of implant materials [1].

AB  - Electrophoretic deposition (EPD) was employed to fabricate composite coatings aimed for orthopedic implants. Hydroxyapatite (HAP), poly(vinyl alcohol) (PVA) and chitosan (CS) coatings, with or without antibiotic gentamicin (Gent) were deposited in a single-step process from aqueous suspension, using EPD. Polymer/mineral composite coatings were formed with the aim to mimic the natural composition of bone tissue, along with improved coatings antibacterial properties, at the same time serving as a drug carrier, allowing local administration of antibiotic. Structural and morphological characterization of HAP/PVA/CS and HAP/PVA/CS/Gent coatings was investigated by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), thermogravimetric analysis (TG/DTG), and scanning electron microscopy (SEM) proving carbonate-substituted HAP. The presence of gentamicin contributed to the bactericidal activity of composite HAP/PVA/CS/Gent coating against Staphylococcus aureus and Escherichia coli. Cytotoxicity studies were performed towards human MRC-5 and mice L929 fibroblast cell lines, and both investigated samples (HAP/PVA/CS and HAP/PVA/CS/Gent) expressed good biocompatibility, without causing toxic effect on tested cells. Biomineralization potential was investigated by measuring the alkaline phosphatase activity, demonstrating high potential of HAP/PVA/CS/Gent to induce the growth of new HAP layer. Therefore, the present investigation proposes the production of composite coatings from non-toxic (aqueous) suspensions, consisting of biocompatible and bioactive components, along with antibiotic gentamicin, improving the antibacterial coatings' properties.

AB  - Biocomposite hydroxyapatite/poly(vinyl alcohol)/chitosan/gentamicin coatings were fabricated on titanium by an electrophoretic deposition process (EPD) from an aqueous suspension using the constant voltage method. Characterization of the deposited coatings was performed by scanning electron micros-copy with field emission (FE-SEM), Fourier transform infrared spectroscopy (FT-IR), and X-ray diffraction (XRD) after immersion in simulated body fluid (SBF) at 37 °C. The concentration of the released gentamicin was determined using high-performance liquid chromatography (HPLC) coupled with a mass spectrometer (MS). The release profile confirmed an initial “burst-release effect” with ~ 30 % of the gen-tamicin released in the first 48 h, which could be of assistance in the prevention of biofilm formation. An-tibacterial activity was proven by the agar diffusion method on Staphylococcus aureus TL and Escherich-ia coli ATCC 25922 bacterial strains. Cytotoxicity was tested by the dye exclusion test (DET) on MRC-5 and L929 fibroblast cell lines, with both coatings expressing nontoxicity. The results showed the high ap-plicability potential of a poly(vinyl alcohol)-based biocomposite coating for medical purposes.

AB  - Medical implants have improved the quality of life of many patients. However, surgical intervention may eventually lead to implant microbial contamination. The aims of this research were to develop an easy, robust, quantitative assay to assess surface antimicrobial activities, especially the anti-nascent biofilm activity, and to identify control surfaces, allowing for international comparisons. Using new antimicrobial assays to assess the inhibition of nascent biofilm during persistent contact or after transient contact with bacteria, we show that the 5 cent Euro coin or other metal-based antibacterial coins can be used as positive controls, as more than 4 log reduction on bacterial survival was observed when using either S. aureus or P. aeruginosa as targets. The methods and controls described here could be useful to develop an easy, flexible and standardizable assay to assess relevant antimicrobial activities of new implant materials developed by industries and academics.

AB  - Recent advances in biomaterials science are increasingly focused
on (re)search for next-generation soft tissue regeneration solutions.
Hydrogel-based materials are becoming a focal point to address
many tissue engineering requirements, possessing exceptional biocompatibility,
with tailorable shape, structure and mechanical
properties, as well as infinite possibilities drug delivery. The widespread
antibiotics use has led to aggravated resistance issues in many
bacterial strains, so the focus is increasingly shifted towards alternative
antibacterial solutions such as silver nanoparticles (AgNPs).
In this work, we have developed novel composite materials based on
poly(vinyl alcohol)/chitosan (PVA/CHI) and poly(vinyl alcohol)/
chitosan/graphene (PVA/CHI/Gr) hydrogels with silver nanoparticles
obtained through green in situ constant-voltage electrochemical
synthesis. The obtained AgNPs’ properties, sizes and size
distributions were examined using UV-visible spectroscopy, dynamic
light scattering and transmission electron microscopy. In vitro
swelling and silver release kinetics were monitored in the simulated
physiological conditions (pH7.4, 37  C). The obtained release and
swelling isotherms were fitted with several theoretical models that
helped discern the mechanisms of these processes. Tensile testing
was carried out to examine the composites’ mechanical behavior and
elasticity – important properties for soft tissue engineering materials.
Finally, the excellent antibacterial activity of the obtained silver/
poly(vinyl alcohol)/chitosan (Ag/PVA/CHI) and silver/poly(vinyl
alcohol)/chitosan/graphene (Ag/PVA/CHI/Gr) hydrogels against
Staphylococcus aureus and Escherichia coli was confirmed quantitatively
by colony counting method, and their non-toxicity was
verified towards two model fibroblast cell lines (MRC-5 and L929)
utilizing the MTT assay. Through thorough characterization, the
obtained nanocomposite hydrogel materials were identified as strong
candidates for active antibacterial soft tissue engineering materials.

AB  - The ethical dilemmas of using animals as in vivo models in preclinical and clinical examinations have been increasingly present in recent decades. Small laboratory animals (rats, rabbits) will continue to be used because they are cost-effective and permit the formation of statistically testable cohort groups; a task that, for financial, maintenance and care reasons, is almost prohibitive for larger animals. Technological advances in the production of new biomaterials for clinical use are enormous, but screening tests and methods used to assess biocompatibility lag behind these advances. The assessment of biological responses is slow and based on millennial recovery mechanisms in eukaryotic organisms. Therefore, the goal of researchers in this field is to re-evaluate old methods of biocompatibility assessment and introduce new methods of evaluation, especially for in vivo testing. In that sense, a revision of the ISO standards was planned and conducted in 2017, which insisted on cytotoxicity testing in cell lines and produced concrete proposals on how biocompatibility should be quantified. In vivo biocompatibility evaluation of biomaterials used for soft tissue recovery commonly utilises rats. Rabbits are recommended for implants used for hard tissues, because of the rabbit’s size, the possibility of implanting the biomaterials on a larger bone surface, and because of the peculiarities of rabbit bone tissue that favours rapid recovery after bone defects and enables easy reading of the results.
AB  - Etičke dileme koje proizilaze iz korišćenja životinja kao in vivo modela u pretkliničkim
i kliničkim ispitivanjima su sve prisutnije poslednjih decenija. Male laboratorijske
životinje će se koristiti i u neposrednoj budućnosti zbog ekonomske isplatljivosti, kao i
zbog toga što omogućavaju formiranje grupa iz kojih se mogu dobiti statistički obradivi
podaci, što nije slučaj sa velikim životinjama. Tehnološki napredak u proizvodnji novih
biomaterijala za kliničku primenu je ogroman, ali skrining testovi i metode koje se
koriste za procenu biokompatibilnosti zaostaju za tim napretkom. Procena biološkog
odgovora nakon implantacije biomaterijala je spora i zasniva se na milenijumskim
mehanizmima oporavka prisutnim kod eukariota. Cilj istraživača iz ove oblasti je
preispitivanje klasičnih metoda procene biokompatibilnosti i uvođenje novih, posebno
za in vivo istraživanja. U cilju ostvarivanja ovog zadatka, u 2017. godini je sprovedena
revizija odgovarajućih ISO standarda, pa se sada insistira na ispitivanju citotoksičnosti
novih biomaterijala na ćelijskim linijama. Takođe, neki istraživači su izneli konkretne
predloge o uspostvaljnju novih načina procenjivanja biokompatibilnosti. U proceni
biokompatibilnosti biomaterijala in vivo koji se koriste za oporavak mekog tkiva,
uobičajeno se koriste pacovi, dok se kunići preporučuju za implantate koji se koriste
za tvrda tkiva zbog njihove veličine i mogućnosti implantacije biomaterijala na veću
površinu kostiju, kao i zbog osobenosti koštanog tkiva kunića koje se brže oporavlja
nakon oštećenja i omogućava lako očitavanje rezultata.

Silver nanoparticles, as a wide-spectrum antibacterial agent that does not induce bacterial resistance, are very convenient for use as an active component in antibacterial hydrogel wound dressing materials with the controlled release ability. In this work, we aim to perform a comprehensive analysis of silver release from poly(vinyl alcohol)-based hydrogels with electrochemically synthesized antibacterial silver nanoparticles (AgNPs), aimed for wound dressing applications. Different AgNPs concentrations were loaded in hydrogel matrices using the green electrochemical reduction method, and thus synthesized nanocomposite hydrogels were characterized by different techniques (scanning electron microscopy, Fourier-transform infrared and Raman spectroscopy, and differential scanning calorimetry), in order to assess their physico-chemical and thermal properties. Silver release was monitored for 28 days in physiologically-relevant conditions (phosphate buffered medium, pH 7.4, 37 degrees C). The obtained experimental data were evaluated against several theoretical release models (Korsmeyer-Peppas, Higuchi, Makoid-Banakar, Kopcha, and early-time approximation), which confirmed the predominantly diffusion-controlled mechanism of release. Finally, AgNP-loaded hydrogels were assessed for antibacterial activity against Staphylococcus aureus TL and Escherichia coli ATCC25922 bacterial strains, to prove their strong potential for wound dressing applications.

Cell-surface interactions stimulate adhesion and activation of macrophages whose acquaintance can assist in designing novel biomaterials that promote favorable macrophage-biomaterial surface interactions for clinical application. This study is designed to determine the distribution and number of macrophages as a means of biocompatibility evaluation of two newly synthesized materials [silver/poly(vinyl alcohol) (Ag/PVA) and silver/poly(vinyl alcohol)/graphene (Ag/PVA/Gr) nanocomposite hydrogels] in vivo, with approval of the Ethics Committee of the Faculty of Veterinary Medicine, University of Belgrade. Macrophages and giant cells were analyzed in tissue sections stained by routine H&E and immunohistochemical methods (CD68(+)). Statistical relevance was determined in the statistical software package SPSS 20 (IBM corp). The results of the study in terms of the number of giant cells localized around the implant showed that their number was highest on the seventh postoperative day (p.o.d.) in the group implanted with Ag/PVA hydrogels, and on the 30th p.o.d. in the group implanted with Ag/PVA/Gr. AB  - Outstanding characteristics of graphene are its high thermal conductivity, inherent high capacity, extremely large specific surface area, high strength, ductility, and remarkable chemical inertness, making it an attractive candidate in the corrosion barrier field. Since graphene coating does not change the thickness and appearance of the substrate, it is an ideal coating for protecting a metal substrate from destructive effects. Between various deposition procedures of graphene coatings on metal surfaces, i.e., electrophoretic deposition, dip coating, spray coating, spin coating, etc., chemical vapour deposition (CVD)-grown graphene coatings have been shown to improve the corrosion resistance of graphene-coated metals significantly. This review is focused on the protective properties of graphene coatings deposited by CVD on different metal substrates and exposed to corrosive environments.

AB  - This paper describes a promising electrophoretic deposition (EPD) approach for developing composite coatings based on chitosan with the additional components of hydroxyapatite, graphene, and gentamicin on titanium substrate. Bioactive properties were investigated in vitro by immersing the coatings in simulated body fluid (SBF) at 37 degrees C. The newly formed biomimetic layer on the top of the deposited chitosan-based coatings on Ti was confirmed by X-ray diffraction, Fourier transform infrared spectroscopy, field emission scanning electron microscopy and electrochemical measurements, while coatings' bioactivity was proved by alkaline phosphatase activity assay in MRC-5 and L929 tested cell lines. The biocompatibility towards MRC-5 and L929 cell lines was investigated by dye exclusion test (DET) implying the non-cytotoxic effect of coatings. Gentamicin release studies were monitored during 21-day by high-performance liquid chromatography coupled with mass spectrometry, indicating rapid release of gentamicin (approximate to 40%) in the first 48 h and more than 60% after 14 days.

AB  - Current trends in biomaterials science address the issue of integrating artificial materials as orthopedic or dental implants with biological materials, e.g., patients' bone tissue. Problems arise due to the simple fact that any surface that promotes biointegration and facilitates osteointegration may also provide a good platform for the rapid growth of bacterial colonies. Infected implant surfaces easily lead to biofilm formation that poses a major healthcare concern since it could have destructive effects and ultimately endanger the patients' life. As of late, research has centered on designing coatings that would eliminate possible infection but neglected to aid bone mineralization. Other strategies yielded surfaces that could promote osseointegration but failed to prevent microbial susceptibility. Needless to say, in order to assure prolonged implant functionality, both coating functions are indispensable and should be addressed simultaneously. This review summarizes progress in designing multifunctional implant coatings that serve as carriers of antibacterial agents with the primary intention of inhibiting bacterial growth on the implant-tissue interface, while still promoting osseointegration.

AB  - A newly produced biomaterial is necessarily subject of standards, which are performed in vivo on animal models. For the evaluation of soft tissue regenerative possibilities after subcutaneous implantation of biomaterials - silver/poly(vinyl alcohol) (Ag/PVA) and novel silver/poly(vinyl alcohol)/graphene (Ag/PVA/Gr) provided for clinical use, sixteen rats were used, according to the instructions of international standards, ISO 10993-6, 2007. Histological sections were observed 7, 15, 30 and 60 days after grafting. These hydrogels were produced by in situ electrochemical synthesis of silver nanoparticles in the polymer matrices, which enabled obtaining completely safe and biocompatible materials, free from any additional toxic chemical reducing agents. Surgical implantation of hydrogels was done according to the permission of the Ethical Committee of the Faculty of Veterinary Medicine, University of Belgrade. Immunohistochemical (IHC) studies included the assessment of smooth muscle expression actin in blood vessels (alpha-SMA), the expression of laminin and type I and type III collagen in the skin structures, and, the determination of cell proliferation marker expression (Ki-67) keratinocytes. The results were assessed in a semiquantitative manner. The data were analyzed in the statistical software package IBM SPSS 20. The conclusions indicated that Ag/PVA/Gr might be used as wound dressings to enhance the tissue healing potential and established faster integration and shorter retention in the tissue.

AB  - Composite bioceramic coatings have been attracting increasing attention of the scientific
public in recent decades due to the good potential of their use for medical purposes. The
main goal of this scientific research is to show the characterization of the obtained
hydroxyapatite (HAP), chitosan (CS) and polyvinyl alcohol (PVA) bioceramic composite
coatings loaded with gentamicin (Gent), on titanium (Ti) substrate using the single-step
electrophoretic deposition process. Titanium, traditionally used for the purpose of replacing
parts of damaged bone tissue, has shown good mechanical properties and biocompatibility
through many years of orthopedic practice. Hydroxyapatite provided the HAP/CS/PVA/Gent
composite coating with osteoinductivity, while the presence of chitosan and polyvinyl
alcohol contributed to its adhesive and mild analgesic properties. The antibacterial agent
gentamicin was added to the HAP/CS/PVA composite, which ensured its direct antibacterial
effect at the implantation site. The coatings were deposited using the cathodic electrophoretic
deposition process, with the Ti plate as the cathode, at a constant voltage of 7V, in the range
of the deposition time from 5 to 12 min. To demonstrate the presumed bioactivity,
HAP/CS/PVA/Gent coatings were immersed in simulated body fluid (SBF) at 37⁰C for
periods of 7 and 14 days. Characterization of HAP/CS/PVA/Gent coatings after immersion
was performed through employment of Fourier transform infrared spectroscopy (FTIR), X ray diffraction (XRD), and field emission scanning electron microscopy (SEM) techniques.
Antibacterial activity was tested by agar diffusion method on two bacterial strains,
Echerichia coli and Staphylococcus aureus with a positive response to both bacterial strains.
The final conclusions of this research suggest a good potential for utilization of
HAP/CS/PVA/Gent bioceramic composite coating, applied through electrophoretic
deposition process on Ti substrate, for medical purposes in the replacement and healing of
damaged bone tissue.

AB  - Polymer-based hydrogel materials are excellent candidates for new-generation wound dressings with improved properties, such as high sorption ability, good mechanical properties and low adhesiveness. Cross-linked hydrogel matrices also serve as excellent carriers for controlled release of antibacterial agents, such as silver nanoparticles (AgNPs), which are preferred over conventional antibiotics due to low propensity to induce bacterial resistance. In this work, we aim to produce novel silver/poly(vinyl alcohol)/chitosan (Ag/PVA/CHI) hydrogels for wound dressing applications. The electrochemical AgNPs synthesis provided facile and green method for the reduction of Ag+ ions inside the hydrogel matrices, without the need to use toxic chemical reducing agents. The formation of AgNPs was confirmed using UV-visible spectroscopy, scanning and transmission electron microscopy. Release kinetics was investigated in modified phosphate buffer solution at 37 degrees C to mimic physiological conditions. Release profiles indicated "burst release" behavior, which is beneficial for wound dressing applications. The antibacterial activity was evaluated against Staphylococcus aureus and Escherichia coli strains using disc-diffusion test, and non-toxicity of hydrogels was proved by dye-exclusion test. The obtained results confirmed strong potential of Ag/PVA/CHI hydrogels for biomedical applications.

AB  - Electrophoretic deposition process (EPD) was successfully used for obtaining graphene (Gr)-reinforced composite coating based on hydroxyapatite (HAP), chitosan (CS), and antibiotic gentamicin (Gent), from aqueous suspension. The deposition process was performed as a single step process at a constant voltage (5 V, deposition time 12 min) on pure titanium foils. The influence of graphene was examined through detailed physicochemical and biological characterization. Fourier transform infrared spectroscopy, field emission scanning electron microscopy, thermogravimetric analysis, X-ray diffraction, Raman, and X-ray photoelectron analyses confirmed the formation of composite HAP/CS/Gr and HAP/CS/Gr/Gent coatings on Ti. Obtained coatings had porous, uniform, fracture-free surfaces, suggesting strong interfacial interaction between HAP, CS, and Gr. Large specific area of graphene enabled strong bonding with chitosan, acting as nanofiller throughout the polymer matrix. Gentamicin addition strongly improved the antibacterial activity of HAP/CS/Gr/Gent coating that was confirmed by antibacterial activity kinetics in suspension and agar diffusion testing, while results indicated more pronounced antibacterial effect againstStaphylococcus aureus(bactericidal, viable cells number reduction  gt 3 logarithmic units) compared toEscherichia coli(bacteriostatic, lt 3 logarithmic units).MTT assay indicated low cytotoxicity (75% cell viability) against MRC-5 and L929 (70% cell viability) tested cell lines, indicating good biocompatibility of HAP/CS/Gr/Gent coating. AB  - The electrophoretic deposition process (EPD) was utilized to produce bioactive hydroxyapatite/chitosan (HAP/CS) and hydroxyapatite/chitosan/gentamicin (HAP/CS/Gent) coatings on titanium. The bioactivity of newly synthesized composite coatings was investigated in the simulated body fluid (SBF) and examined by X-ray diffraction, Fourier transform infrared spectroscopy, and field emission scanning electron microscopy. The obtained results revealed carbonate-substituted hydroxyapatite after immersion in SBF, emphasizing the similarity of the biomimetically grown HAP with the naturally occurring apatite in the bone. The formation of biomimetic HAP was confirmed by electrochemical impedance spectroscopy and polarization measurements, through the decrease in corrosion current density and coating capacitance values after 28-day immersion in SBF. The osseointegration ability was further validated by measuring the alkaline phosphatase activity (ALP) indicating the favorable osseopromotive properties of deposited coatings (significant increase in ALP levels for both HAP/CS (3.206 U mL(-1)) and HAP/CS/Gent (4.039 U mL(-1)) coatings, compared to the control (0.900 U mL(-1))). Drug-release kinetics was investigated in deionized water at 37 degrees C by high-performance liquid chromatography coupled with mass spectrometry. Release profiles revealed the beneficial "burst-release effect" (similar to 21% of gentamicin released in the first 48 h) as a potentially promising solution against the biofilm formation in the initial period. When tested against human and mice fibroblast cells (MRC-5 and L929), both composite coatings showed a noncytotoxic effect (viability  gt 85%), providing a promising basis for further medical application trials.

AB  - This study reports in-situ cesium-doped graphene (G/Cs) coating obtained by introducing Cs2CO3 into the atmospheric pressure chemical vapor deposition (APCVD) furnace during graphene deposition on copper. The successful Cs-doping of the graphene coating was confirmed via X-ray photoelectron spectroscopy (XPS). As compared to the spectra of pure graphene coating, the XPS spectra of the G/Cs coating revealed a shift of the C1s and Cs3d(5/2) peaks to higher and lower binding energies, respectively; thus, implying the n-type character of the doping and indicating a charge transfer between Cs and graphene. Raman results show that a pure graphene coating is composed of fewer layers, fewer defects, and larger domain size than the G/Cs coating. Ultraviolet photoelectron spectroscopy was utilized to study the work function of graphene and the G/Cs and revealed that doping graphene with Cs dopants reduced the work function of graphene by 1.2 eV. Electrochemical testing during 15-day immersion in 0.1 M NaCl indicated the destructive effect of the G/Cs coating on the Cu substrate. The results showed that the G/Cs coating exhibits a higher corrosion rate and lower corrosion resistance than even the bare metal itself.

Lead isotope ratios as tool for elucidation of chemical environment in a system of Macrolepiota procera (Scop.) Singer - soil

AB  - The analysis of isotope ratios of lead in the mushrooms and soil, where they were grown, assisted with a principal component analysis, offered a new perspective for understanding possible chemical environment in a real setup of those compartments. The content of lead and its isotope compositions were determined in soil samples and mushroom Macrolepiota procera from unpolluted area of Mountain Goč, Serbia. Sequential extraction procedure based on the Commission of the European Community Bureau of Reference (BCR) was applied on soil samples in order to determine the distribution of lead in the labile and un-labile fractions of the soil. Caps and stipes of mushrooms were subjected to microwave acid-assisted digestion prior to measurements by inductively coupled plasma quadrupole mass spectrometer for determination of lead content and lead isotope ratios. Information about the chemical fractionation of Pb in soil, Pb isotopic data from soil fractions and fruiting bodies allowed a more detailed insight on the uptake mechanisms. Lead was predominantly associated with reducible fraction (~ 60%). Only its small portion (∼ 1%) was present in the exchangeable and acid-extractable fractions suggesting the low mobility of Pb. Lead isotope analysis revealed the presence of anthropogenic lead in the surface soil. Significant lower 206Pb/207Pb compared with other fractions was found in exchangeable and acid-soluble fraction (1.331 ± 0.010), which corresponds to the isotope ratio of European gasoline. The highest 206Pb/207Pb ratio was observed in reducible fraction (1.162 ± 0.007), while in oxidizable and residual fraction, those values were similar (1.159 ± 0.006 and 1.159 ± 0.004, respectively). Distinction of exchangeable and acid-extractable fractions from others was also confirmed, for the first time, by principal component analysis. The analysis of four isotope ratios (206Pb/207Pb, 208Pb/206Pb, 206Pb/204Pb, and 207Pb/204Pb) indicated that the analyzed M. procera accumulates lead from the first two fractions of topsoil layers.

AB  - Modernization and improvement of wound dressing materials is an important topic in biomaterials and biomedicine fields, as the traditional materials are inadequate and susceptible to bacterial infections. In recent times, polymer-based hydrogel materials have presented themselves as excellent candidates for new-generation wound dressings with improved properties, such as high sorption ability, good mechanical properties, and low adhesiveness. Additionally, cross linked hydrogel matrices serve as excellent carriers for controlled release of antibacterial agents, such as silver nanoparticles (AgNPs), which are preferred over conventional antibiotics due to multi-phase mechanism of action and low susceptibility to induce bacterial resistance. Their incorporation inside polymer matrices allows improvement of wound dressing properties and sustained protection against bacterial infection. Electrochemical methods for AgNPs synthesis are facile and green alternatives to chemical routes, allowing the formation of highly stable AgNPs with strong antibacterial effect. In this article, we aim to provide a comprehensive review of the existing research on the topic of electrochemically synthesized silver nanoparticles incorporated in polymer matrices with a special focus on the chitosan-based hydrogels as prospective materials for wound dressing applications.

AB  - Silver nanoparticles (AgNPs) have become an interesting alternative to antibiotics for potential applications in biomedicine, since nanocrystalline silver is proved to be highly efficient antimicrobial agent with a wide inhibiting spectrum against many types of microorganisms. AgNPs embedded in hydrogel matrices are attractive for biomedical applications due to the possibility for their controlled release resulting in enhanced antimicrobial activity. Thus, combination of AgNPs with biocompatible hydrogels, poly(vinyl alcohol) (PVA) and chitosan (CHI), provides potential for design of improved medical treatments and devices (antimicrobial wound dressings, soft tissue implants). Graphene (Gr) has exceptional mechanical properties and has therefore been applied as adequate reinforcing component for polymer-based composite materials. The synthesis of AgNPs is always subject to innovation and improvements, as it is desirable to minimize the use of chemical agents with the aim of improving the biocompatibility of the obtained material. With this in mind, a green alternative to traditional chemical synthesis methods would be the electrochemical reduction of silver ions, especially as this method provides the option of direct in situ synthesis of AgNPs inside the polymer matrix. In this paper, we provide an overview of the state of the art regarding graphene-reinforced polymer matrices with electrochemically synthesized silver nanoparticles for medical application. We show that the electrochemical method of AgNPs synthesis in PVA/CHI hydrogel matrices has emerged as an excellent method to obtain nanoparticles, and that the incorporation of graphene improved the hydrogels` mechanical properties and elasticity.

The physicochemical properties of nanocomposite hydrogels were investigated by UV-vis spectroscopy (UV-vis), dynamic light scattering (DLS), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), field-emission scanning electron microscopy (FE-SEM) and Fourier transform infrared spectroscopy (FT-IR), thermal characteristics were determined by differential scanning calorimetry (DSC) and mechanical properties were measured by tensile test. The swelling studies were carried out in phosphate buffer to simulate natural physiological environment and data were fitted by several kinetic models to determine the diffusion mechanism and diffusion coefficients of the swelling medium through the hydrogel matrices. It was shown that the presence of silver nanoparticles increased the uptake capability and equilibrium swelling degree of the composite hydrogels. The antibacterial activity was confirmed against Escherichia coli and Staphylococcus aureus, while the hydrogels without AgNPs exhibited antibacterial properties due to the presence of chitosan. With the addition of AgNPs, the samples showed stronger activity and fast reduction in the number of colonies, confirming the synergistic effect of chitosan and AgNPs on the antibacterial activity.

The swelling studies were carried out in phosphate buffer to simulate natural physiological environment and data were fitted by several kinetic models to determine the diffusion mechanism and diffusion coefficients of the swelling medium through the hydrogel matrices. It was shown that the presence of silver nanoparticles increased the uptake capability and equilibrium swelling degree of the composite hydrogels. The antibacterial activity was confirmed against Escherichia coli and Staphylococcus aureus, while the hydrogels without AgNPs exhibited antibacterial properties due to the presence of chitosan. With the addition of AgNPs, the samples showed stronger activity and fast reduction in the number of colonies, confirming the synergistic effect of chitosan and AgNPs on the antibacterial activity.",

AB  - In recent times, polymer-based hydrogel materials have presented themselves as excellent candidates for
new-generation wound dressings with improved properties, such as high sorption ability, good mechanical
properties and low adhesiveness [1]. Additionally, cross linked hydrogel matrices serve as excellent carriers
for controlled release of antibacterial agents, such as silver nanoparticles (Ag/NPs), which are preferred over
conventional antibiotics due to multi-phase mechanism of action and low susceptibility to induce bacterial
resistance [2]. In this work, we aim to produce novel silver/poly(vinyl alcohol)/chitosan (Ag/PVA/CHI)
hydrogels for wound dressing applications. The electrochemical route for AgNPs synthesis provided facile
and green method for the reduction of Ag+
ions inside the hydrogel matrices, without the need to use toxic
chemical reducing agents [3]. The effect of chitosan content on the synthesis yield, antibacterial properties,
swelling and release kinetics was investigated. The formation of AgNPs was confirmed using UV-visible
spectroscopy through the appearance of plasmon resonant peaks at around 400 nm (Figure 1a), whereas
transmission electron microscopy (TEM) proved the incorporation of both single and polycrystalline spherical
AgNPs with diameters of 5-10 nm (Figure 1b). The swelling and silver release kinetics were investigated in
modified phosphate buffer solutions (pH 7.4) at 37 °C to mimic physiological conditions. The obtained
swelling isotherms and release profiles were fitted with different models to obtain kinetic and diffusion
parameters (Figure 1c). The antibacterial activity was evaluated against Staphylococcus aureus TL and
Escherichia coli ATCC 25922 bacterial strains using an in-suspension test, and non-toxicity of both silvercontaining and silver-free hydrogels was proved by MTT cytotoxicity test. The obtained results confirmed
strong potential of Ag/PVA/CHI hydrogels for biomedical applications.

AB  - In this work, we provide a facile path for the preparation of wound dressing materials with improved characteristics based on biocompatible chitosan and poly(vinyl alcohol) hydrogels with embedded silver nanoparticles (AgNPs) as a potent antimicrobial agent. The hydrogels were prepared via freezing-thawing method, whereas AgNPs were synthesized via an in situ electrochemical route. The excellent physicochemical properties of the obtained nanocomposite hydrogels were validated using different spectroscopic and microscopic techniques. The formation of sub-10 nm AgNPs was confirmed by UV-visible spectroscopy, dynamic light scattering and transmission electron microscopy. Swelling and silver release profiles were validated using several diffusion models. The hydrogels were proven to be non-cytotoxic using MTT test and possessed powerful antibacterial activity against Staphylococcus aureus and Escherichia coli, as determined by test in suspension, which affirmed their feasibility for wound dressing applications.
